BY DEMONSTRATING THE INTEGRATION OF MULTIPLE DECARBONIZATION PATHWAYS, THIS PROJECT SEEKS TO HELP A MAJOR AMERICAN BRAND ACHIEVE DEEP DECARBONIZATION AND SERVE AS AN EXAMPLE FOR OTHER U.S. FOOD AND BEVERAGE COMPANIES TO REDUCE EMISSIONS FROM PROCESS HEAT WHILE REDUCING ENERGY COSTS. THIS PROJECT EXPECTS TO CREATE AN ESTIMATED 500 CONSTRUCTION JOBS ACROSS THE 10 SITES. THREE OF THE 10 PROJECT SITES (FREMONT, OH; HOLLAND, MI; AND MUSCATINE, IA) CURRENTLY HAVE UNIONIZED LABOR. ALL SITES HAVE PEOPLE COMMITTEES AS PART OF THE KRAFT HEINZ MANAGEMENT SYSTEM THAT INCLUDES HOURLY AND SALARIED EMPLOYEE REPRESENTATIVES INCLUDING HUMAN RESOURCES. AN ENERGY CHAMPION WILL JOIN THE COMMITTEE TO REPRESENT ANY WORKER CONCERNS AND SERVE AS COORDINATOR BETWEEN THE PROJECT TEAM AND PLANT WORKERS. | Department of Commerce | PURPOSE: THE PROPOSED PROJECT WILL DEVELOP THE 222-MILE COUNTY OF GRAFTON MIDDLE-MILE BACKBONE NETWORK THROUGH POPULATION CENTERS THROUGH A COMBINATION OF BUILDING NEW FIBER OPTIC CABLE WHERE THERE ARE OBVIOUS GAPS AND OPTIMIZING EXISTING INFRASTRUCTURE WHERE POSSIBLE, REDUCING COSTS OF THE BUILDOUT. THE BACKBONE WILL BE AVAILABLE FOR MUNICIPALITIES TO THEN TAP INTO AND RUN ADDITIONAL FIBER OPTIC CABLE THROUGH THEIR MUNICIPALITIES TO REACH THE LAST MILE. THE NETWORK WILL PROVIDE THE ELECTRONICS AND OPTICS NECESSARY TO PROVIDE SERVICE AT IDENTIFIED MUNICIPAL/COUNTY ANCHOR LOCATIONS AND WILL ALLOW FOR FUTURE GROWTH AND EXPANSION WITH MINIMAL ADDITIONAL EQUIPMENT. THE NON-MUNICIPAL/COUNTY LOCATIONS WILL BE SERVED THROUGH DARK FIBER ACCESS ONLY, ALLOWING ISPS TO MAKE THEIR OWN INVESTMENTS WITHIN INDIVIDUAL COMMUNITIES. THE RURAL NATURE OF THE REGION, PAIRED WITH THE FACT THAT THERE ARE LESS THAN 53 PERSONS PER SQUARE MILE, PREVENTS PRIVATE INTERNET SERVICE PROVIDERS FROM MAKING THE SIGNIFICANT UPFRONT INVESTMENT TO DEVELOP A MIDDLE-MILE OR LAST MILE BROADBAND NETWORK. BY CENTRALIZING THE MIDDLE MILE BACKBONE, A GREAT NUMBER OF INDIVIDUALS WILL BE SERVED WITHIN A SHORT TIME FRAME, BRINGING SPEEDS OF UP TO 1 GBPS TO THE REGION. THE PROJECT WILL ENCOURAGE THE EXPANSION OF MIDDLE MILE INFRASTRUCTURE TO REDUCE THE COST OF CONNECTING UNSERVED AND UNDERSERVED AREAS TO THE BACKBONE OF THE INTERNET AND WILL PROMOTE BROADBAND CONNECTION RESILIENCY THROUGH THE CREATION OF ALTERNATIVE NETWORK CONNECTION PATHS THAT ARE DESIGNED TO PREVENT SINGLE POINTS OF FAILURE ON THE BROADBAND NETWORK.ACTIVITIES TO BE PERFORMED AND EXPECTED OUTCOMES: (1.) DEPLOY A COUNTYWIDE OPEN-ACCESS, MIDDLE-MILE, FIBER-OPTIC NETWORK THROUGHOUT THE COUNTY WITH DARK FIBER ACCESS SERVICE.(2.) FACILITATE INTERCONNECTIONS TO DARTMOUTH COLLEGE AND THE UNIVERSITY OF NEW HAMPSHIRE TO SUPPORT EDUCATION AS WELL AS PROVIDE ADDITIONAL REDUNDANCY AND NETWORK RESILIENCY.(3.) SERVE ALL THE MUNICIPALITIES WITHIN THE COUNTY WITH A DARK FIBER BACKBONE THAT IS CAPABLE OF SUPPORTING LAST MILE SPEEDS WITH A MINIMUM OF 100MBPS UP TO A 1GBPS TO HOMES AND BUSINESSES.INTENDED BENEFICIARIES: WITHIN THE PROJECT AREA, THERE ARE AN ESTIMATED 6,100 HOUSEHOLDS CONSIDERED UNSERVED BY NTIA DEFINITION (12% OF THE TOTAL). ACCORDING TO BROADBANDNOW.COM/NEW-HAMPSHIRE, OF THE 10 COUNTIES IN THE STATE, GRAFTON COUNTY IS RANKED AS THE 4TH COUNTY IN THE STATE WITH THE LEAST AVAILABILITY OF 100 MBPS BROADBAND SERVICE THIS PROJECT WILL POISE THE REGION TO DELIVER CRITICAL BROADBAND SERVICES TO THESE HOUSEHOLDS.WITHIN THE COUNTY THERE ARE 27 K-12 EDUCATION FACILITIES, 41 LICENSED CHILDCARE FACILITIES, 5 HIGHER-EDUCATION INSTITUTIONS, 2 CAREER TECHNOLOGY CENTERS, AND 5 HOSPITALS. THE BUILDOUT OF THE MIDDLE-MILE GRAFTON COUNTY BROADBAND NETWORK WILL ALLOW THESE ANCHOR INSTITUTIONS TO ACCESS AFFORDABLE, HIGH-SPEED INTERNET SERVICES TO SUPPORT THEIR RESIDENTS AND WORKFORCE.THIS NETWORK INFRASTRUCTURE WILL PROVIDE THE COUNTY, ITS RESIDENTS, AND BUSINESSES THE OPPORTUNITY TO ASCERTAIN QUALITY BROADBAND SERVICES AT LEVELS THAT DO NOT EXIST TODAY. ASPECTS OF SEVERE ASTHMA CURRENTLY NOT MANAGED BY AVAILABLE THERAPIES MAY DISPLAY A STRONG INFLAMMATORY COMPONENT DRIVEN BY THE INNATE AND ADAPTIVE IMMUNE SYSTEMS, PARTICULARLY FOLLOWING VIRAL EXACERBATIONS. DEVELOPMENT OF NEW THERAPEUTIC STRATEGIES TARGETING A WIDE RANGE OF INFLAMMATORY MECHANISMS, NOT ONLY TH2 AND TH1/TH17 RESPONSES, IS NEEDED TO CONTROL SEVERE ASTHMA. RAFT PHARMACEUTICALS PROPOSES A NEW TARGET FOR TREATMENT OF SEVERE ASTHMA – OVERABUNDANT AND CLUSTERED, PATHOLOGICAL LIPID RAFTS IN BRONCHIAL EPITHELIAL AND IMMUNE CELLS IN ASTHMATICS. CHOLESTEROL-RICH LIPID RAFTS PROVIDE ORDERED PLASMA MEMBRANE DOMAINS, WHERE RECEPTORS, ION CHANNELS AND ADAPTOR MOLECULES CAN ASSOCIATE TO FORM FUNCTIONAL COMPLEXES. LARGE LIPID RAFTS ARE REQUIRED AS THE LANDING PAD FOR MICROBES ON HOST CELLS AND FOR INITIATION OF MANY INFLAMMATORY PROCESSES IN BRONCHIAL EPITHELIAL AND IMMUNE CELLS. WE DISCOVERED APOA-I BINDING PROTEIN (AIBP) AS A MOLECULE THAT SELECTIVELY TARGETS THE CHOLESTEROL DEPLETION MACHINERY TO PATHOLOGIC LIPID RAFTS IN INFLAMMATORY CELLS, WITHOUT AFFECTING HOMEOSTATIC CELLULAR FUNCTION. THE LATTER EXPLAINS AN EXCEPTIONAL SAFETY PROFILE OF AIBP IN MICE AND RATS. IN THE PHASE I OF THIS GRANT, USING MOUSE MODELS OF ASTHMA, WE DEMONSTRATED THAT ADMINISTRATION OF A BIOLOGIC DERIVED FROM THE AIBP PROTEIN, WAS EFFECTIVE IN AN ACUTE HDM MODEL OF ASTHMA IN MICE, REDUCING AIRWAY HYPERRESPONSIVENESS, AIRWAY EOSINOPHILIA, AND EXPRESSION OF TH2 CYTOKINES AND EPITHELIAL ALARMINS. IN A MODEL OF SEVERE ASTHMA, AIBP SIGNIFICANTLY REDUCED PULMONARY NEUTROPHILIA. FOR PHASE II PROPOSAL, A TEAM OF EXPERTS IN BIOLOGIC DRUG DEVELOPMENT AND IN PRECLINICAL ASSESSMENT AND CLINICAL TRIALS OF THERAPEUTIC AGENTS IN ASTHMA HAS BEEN ASSEMBLED TO FURTHER THE CHARACTERIZATION OF AN IMPROVED AIBP SEQUENCE, RFT1124, AS A DEVELOPMENT CANDIDATE FOR SEVERE ASTHMA. SPECIFICALLY, WE PLAN TO MANUFACTURE AND RELEASE RFT1124 DRUG PRODUCT FOR EFFICACY AND TOXICOLOGY STUDIES USING CGMP-COMPATIBLE PROCESSES AND ANALYTICAL ASSAYS. THE EFFICACY OF RFT1124 AS AN ADD-ON THERAPY TO STANDARD-OF-CARE INHALED CORTICOSTEROIDS AND LONG-ACTING BETA-AGONISTS (ICS+LABA) WILL BE TESTED IN MOUSE MODELS OF ASTHMA EXACERBATED BY RHINOVIRUS (RV) INFECTION. IN ADDITION, WE WILL USE PRECISION CUT LUNG SLICES FROM ASTHMATICS AND NON-ASTHMATICS POSTMORTEM LUNG TISSUE TO TEST THE EFFICACY OF ADD-ON RFT1124 IN ICS+LABA TREATMENT IN RV INFECTED HUMAN AIRWAYS. RNASEQ STUDIES WILL ESTABLISH WHETHER RFT1124 INHIBITS CORTICOSTEROID INSENSITIVE ANTI-INFLAMMATORY PATHWAYS AND/OR ENHANCES CORTICOSTEROID SENSITIVE PATHWAYS IN RV INFECTED HUMAN BRONCHIAL EPITHELIAL CELLS. THROUGH ITS EXPERT LEGAL AND SOCIAL SERVICES STAFF, THE COALITION TO ABOLISH SLAVERY AND TRAFFICKING (CAST) PROPOSES TO CONTINUE ITS COMPREHENSIVE, NATIONAL, ACCESSIBLE AND INCLUSIVE TRAINING AND TECHNICAL ASSISTANCE (TTA) PROJECT TO STRENGTHEN LEGAL SERVICE PROVIDERS, PRO BONO ATTORNEYS, AND SOCIAL SERVICE PROVIDERS CAPACITY TO IDENTIFY AND PROVIDE COMPREHENSIVE LEGAL SERVICES TO VICTIMS OF HUMAN TRAFFICKING THROUGH A TRAUMA-INFORMED AND RACE EQUITY LENS. IN PARTNERSHIP WITH THE OFFICE FOR VICTIMS OF CRIME, CAST WILL BUILD ON OVER 15 YEARS OF PROVIDING TTA TO DEEPEN AND EXPAND ITS NATIONAL TTA EFFORTS TO INCREASE TRAFFICKING VICTIMS ACCESS TO QUALITY COMPREHENSIVE LEGAL SERVICES. ADDITIONALLY, CAST WILL LEVERAGE COLLABORATION WITH MANY NATIONAL PARTNERS LIKE FREEDOM NETWORK USAS SURVIVOR REENTRY PROJECT TO PROVIDE COMPREHENSIVE TTA. THE PROJECT WILL PROVIDE COMPREHENSIVE LIVE AND ON-DEMAND TRAINING TO LEGAL AND SOCIAL SERVICE PROVIDERS INCLUDING ANTI-TRAFFICKING LAWYERING TRAINING MODULE 2, INTENSIVE T VISA DECLARATION WORKSHOPS, AND VICTIMS RIGHTS ADVOCACY FOR SOCIAL SERVICE PROVIDERS, AS WELL AS SELF-PACED ONLINE TRAINING TO MANY MORE ATTORNEYS AND SOCIAL SERVICE PROVIDERS NATIONALLY THROUGH AN ADVANCED LEARNING MANAGEMENT SYSTEM FOR ASYNCHRONOUS AND SYNCHRONOUS LEARNING. THE USE OF LIQUID CHROMATOGRAPHY COUPLED WITH MASS SPECTROMETRY (LC-MS) HAS RISEN IN PROMINENCE IN THE FIELD OF METABOLOMICS DUE TO ITS ABILITY TO ANALYZE A SIZABLE NUMBER OF METABOLITES WITH A LIMITED AMOUNT OF BIOLOGICAL MATERIAL. HOWEVER, IN A TYPICAL UNTARGETED METABOLOMICS ANALYSIS OF HUMAN SAMPLES BY LC-MS, ABOUT 70% OF THE DETECTED PEAKS REPRESENT UNKNOWN ANALYTES MAINLY BECAUSE EXISTING MASS SPECTRAL LIBRARIES COVER ONLY A SMALL FRACTION OF KNOWN COMPOUNDS, BUT ALSO DUE TO UNCERTAINTY IN PEAK PICKING, ALIGNMENT OF PEAKS, AND RECOGNIZING ISOTOPIC PEAKS AND ADDUCT FORMS. THESE CHALLENGES HAVE KEPT AT BAY THE PACE OF DEVELOPMENT OF DATA ANALYTICS PIPELINES FOR METABOLOMICS AND ITS INTEGRATION WITH OTHER OMICS STUDIES. THE GOAL OF THIS PHASE II SBIR PROPOSAL IS TO MAKE METABOLOMICS STUDIES ON A PAR WITH OTHER OMICS STUDIES SUCH AS GENOMICS, TRANSCRIPTOMICS, AND PROTEOMICS, FOR WHICH WELL-ESTABLISHED PIPELINES ARE AVAILABLE. BY DOING SO, WE WILL ACCELERATE THE ROLE OF METABOLOMICS IN SYSTEMS BIOLOGY APPROACHES FOR VARIOUS APPLICATIONS INCLUDING BIOMARKER AND DRUG DISCOVERY. TO ACHIEVE THIS GOAL, WE PROPOSE TO DEVELOP A CLOUD-BASED PLATFORM THAT ALLOWS CUSTOMERS TO BUILD PIPELINES FOR ANALYSIS OF LC-MS-BASED UNTARGETED METABOLOMICS DATA, STARTING FROM PEAK DETECTION TO METABOLITE ANNOTATION. THIS WILL BE ACCOMPLISHED BY IMPLEMENTING A SUITE OF INNOVATIVE TOOLS THAT CAN BE ASSEMBLED INTO CUSTOMIZED PIPELINES AND BY ENHANCING METABOLITE ANNOTATION ACCURACY THROUGH INTEGRATION OF INFORMATION DERIVED FROM MULTIPLE RESOURCES INCLUDING COMPOUND DATABASES, PATHWAYS, BIOCHEMICAL NETWORKS, AND MASS SPECTRAL LIBRARIES. AIM 1 OF THIS PROPOSAL WILL FOCUS ON DEVELOPING A SUITE OF TOOLS TO ENABLE: (1) PEAK DETECTION, ALIGNMENT, AND QUALITY ASSESSMENT; (2) ADDUCT AND ISOTOPIC PEAK RECOGNITION; (3) MASS-BASED SEARCH AGAINST MULTIPLE COMPOUND DATABASES; (4) EXPERT-BASED EVALUATION OF PUTATIVE IDS; (5) ISOTOPIC PATTERN ANALYSIS; (6) NETWORK-BASED EVALUATION OF PUTATIVE IDS; (7) SPECTRAL MATCHING OF MS/MS DATA AGAINST EXPERIMENTAL AND IN- SILICO FRAGMENTATION PATTERNS; (8) DEEP LEARNING-BASED PREDICTION OF COMPOUND FINGERPRINTS; AND (9) INTEGRATIVE ASSESSMENT OF PUTATIVE METABOLITE IDS VIA A PROBABILISTIC MODEL. AIM 2 WILL ASSEMBLE THE TOOLS DEVELOPED IN AIM 1 INTO A CLOUD-BASED PLATFORM, METABOQUEST, WHICH PROVIDES USERS WITH INTERACTIVE VISUALIZATION OF PEAKS, ISOTOPIC PATTERNS, NETWORKS, AND MASS SPECTRA. FURTHERMORE, AIM 2 WILL FOCUS ON INTEGRATING INTO METABOQUEST A PIPELINE BUILDER THAT ALLOWS USERS TO CREATE PIPELINES BY LINKING MODULES AND RUN THEM REMOTELY THROUGH A MODULAR INTERACTIVE WEB INTERFACE. AIM 3 WILL PERFORM A COMPREHENSIVE EVALUATION OF METABOQUEST IN TERMS OF METABOLITE ANNOTATION ACCURACY, NUMBER OF ANNOTATED METABOLITES, AND COMPUTATIONAL EFFICIENCY COMPARED TO OTHER EXISTING TOOLS. THE DELICIOUS DECARBONIZATION THROUGH INTEGRATED ELECTRIFICATION AND ENERGY STORAGE PROJECT, LED BY KRAFT HEINZ, PLANS TO UPGRADE, ELECTRIFY, AND DECARBONIZE ITS PROCESS HEAT AT 10 FACILITIES BY APPLYING A RANGE OF TECHNOLOGIES INCLUDING HEAT PUMPS, ELECTRIC HEATERS, AND ELECTRIC BOILERS IN COMBINATION WITH BIOGAS BOILERS, SOLAR THERMAL, SOLAR PHOTOVOLTAIC, AND THERMAL ENERGY STORAGE. THE TAILORED APPLICATION OF THESE TECHNOLOGIES AT EACH FACILITY EXPECTS TO REDUCE ANNUAL EMISSIONS BY MORE THAN 300,000 METRIC TONS OF CARBON DIOXIDE PER YEAR, TRANSLATING TO A 99% REDUCTION FROM 2022 LEVELS. BY DEMONSTRATING THE INTEGRATION OF MULTIPLE DECARBONIZATION PATHWAYS, THIS PROJECT SEEKS TO HELP A MAJOR AMERICAN BRAND ACHIEVE DEEP DECARBONIZATION AND SERVE AS AN EXAMPLE FOR OTHER U.S. FOOD AND BEVERAGE COMPANIES TO REDUCE EMISSIONS FROM PROCESS HEAT WHILE REDUCING ENERGY COSTS. PURPOSE: THE PROPOSED PROJECT WILL DEVELOP THE 222-MILE COUNTY OF GRAFTON MIDDLE-MILE BACKBONE NETWORK THROUGH POPULATION CENTERS THROUGH A COMBINATION OF BUILDING NEW FIBER OPTIC CABLE WHERE THERE ARE OBVIOUS GAPS AND OPTIMIZING EXISTING INFRASTRUCTURE WHERE POSSIBLE, REDUCING COSTS OF THE BUILDOUT. THE BACKBONE WILL BE AVAILABLE FOR MUNICIPALITIES TO THEN TAP INTO AND RUN ADDITIONAL FIBER OPTIC CABLE THROUGH THEIR MUNICIPALITIES TO REACH THE LAST MILE. THE NETWORK WILL PROVIDE THE ELECTRONICS AND OPTICS NECESSARY TO PROVIDE SERVICE AT IDENTIFIED MUNICIPAL/COUNTY ANCHOR LOCATIONS AND WILL ALLOW FOR FUTURE GROWTH AND EXPANSION WITH MINIMAL ADDITIONAL EQUIPMENT. THE NON-MUNICIPAL/COUNTY LOCATIONS WILL BE SERVED THROUGH DARK FIBER ACCESS ONLY, ALLOWING ISPS TO MAKE THEIR OWN INVESTMENTS WITHIN INDIVIDUAL COMMUNITIES. THE RURAL NATURE OF THE REGION, PAIRED WITH THE FACT THAT THERE ARE LESS THAN 53 PERSONS PER SQUARE MILE, PREVENTS PRIVATE INTERNET SERVICE PROVIDERS FROM MAKING THE SIGNIFICANT UPFRONT INVESTMENT TO DEVELOP A MIDDLE-MILE OR LAST MILE BROADBAND NETWORK. BY CENTRALIZING THE MIDDLE MILE BACKBONE, A GREAT NUMBER OF INDIVIDUALS WILL BE SERVED WITHIN A SHORT TIME FRAME, BRINGING SPEEDS OF UP TO 1 GBPS TO THE REGION. THE PROJECT WILL ENCOURAGE THE EXPANSION OF MIDDLE MILE INFRASTRUCTURE TO REDUCE THE COST OF CONNECTING UNSERVED AND UNDERSERVED AREAS TO THE BACKBONE OF THE INTERNET AND WILL PROMOTE BROADBAND CONNECTION RESILIENCY THROUGH THE CREATION OF ALTERNATIVE NETWORK CONNECTION PATHS THAT ARE DESIGNED TO PREVENT SINGLE POINTS OF FAILURE ON THE BROADBAND NETWORK.ACTIVITIES TO BE PERFORMED AND EXPECTED OUTCOMES: (1.) DEPLOY A COUNTYWIDE OPEN-ACCESS, MIDDLE-MILE, FIBER-OPTIC NETWORK THROUGHOUT THE COUNTY WITH DARK FIBER ACCESS SERVICE.(2.) FACILITATE INTERCONNECTIONS TO DARTMOUTH COLLEGE AND THE UNIVERSITY OF NEW HAMPSHIRE TO SUPPORT EDUCATION AS WELL AS PROVIDE ADDITIONAL REDUNDANCY AND NETWORK RESILIENCY.(3.) SERVE ALL THE MUNICIPALITIES WITHIN THE COUNTY WITH A DARK FIBER BACKBONE THAT IS CAPABLE OF SUPPORTING LAST MILE SPEEDS WITH A MINIMUM OF 100MBPS UP TO A 1GBPS TO HOMES AND BUSINESSES.INTENDED BENEFICIARIES: WITHIN THE PROJECT AREA, THERE ARE AN ESTIMATED 6,100 HOUSEHOLDS CONSIDERED UNSERVED BY NTIA DEFINITION (12% OF THE TOTAL). ACCORDING TO BROADBANDNOW.COM/NEW-HAMPSHIRE, OF THE 10 COUNTIES IN THE STATE, GRAFTON COUNTY IS RANKED AS THE 4TH COUNTY IN THE STATE WITH THE LEAST AVAILABILITY OF 100 MBPS BROADBAND SERVICE THIS PROJECT WILL POISE THE REGION TO DELIVER CRITICAL BROADBAND SERVICES TO THESE HOUSEHOLDS.WITHIN THE COUNTY THERE ARE 27 K-12 EDUCATION FACILITIES, 41 LICENSED CHILDCARE FACILITIES, 5 HIGHER-EDUCATION INSTITUTIONS, 2 CAREER TECHNOLOGY CENTERS, AND 5 HOSPITALS. THE BUILDOUT OF THE MIDDLE-MILE GRAFTON COUNTY BROADBAND NETWORK WILL ALLOW THESE ANCHOR INSTITUTIONS TO ACCESS AFFORDABLE, HIGH-SPEED INTERNET SERVICES TO SUPPORT THEIR RESIDENTS AND WORKFORCE.THIS NETWORK INFRASTRUCTURE WILL PROVIDE THE COUNTY, ITS RESIDENTS, AND BUSINESSES THE OPPORTUNITY TO ASCERTAIN QUALITY BROADBAND SERVICES AT LEVELS THAT DO NOT EXIST TODAY. THE EXPANSION OF RELIABLE, MODERN FIBER-OPTIC BROADBAND INFRASTRUCTURE WITHIN THE REGION WILL SPUR ECONOMIC DEVELOPMENT AND PROVIDE THE COUNTY THE SCALABILITY FOR FUTURE SERVICES, INCLUDING PUBLIC SAFETY, EDUCATION, HEALTHCARE AND OTHER GOVERNMENT SERVICES.SUBRECIPIENT ACTIVITIES: DESIGN, PERMITTING, MAKE-READY, CONSTRUCTION/CONSTRUCTION MANAGEMENT, AND MIDDLE MILE NETWORK MANAGEMENT ACTIVITIES WILL BE SUB-AWARDED TO EX2 TECHNOLOGY LLC.

TARGETING LIPID RAFTS FOR TREATMENT OF ASTHMA - PROJECT SUMMARY PATIENTS WITH SEVERE ASTHMA WHO DISPLAY PERSISTENT BRONCHOCONSTRICTION AND/OR REDUCED LUNG FUNCTION DESPITE AVAILABLE BRONCHODILATOR, CORTICOSTEROID AND TH2-TARGETING THERAPY DEVELOP SIGNIFICANT MORBIDITY, WHICH SEVERELY IMPACTS THEIR QUALITY OF LIFE OWING TO PERSISTENT SYMPTOMS, AS WELL AS FREQUENT AND LIFE-THREATENING EXACERBATIONS. ASPECTS OF SEVERE ASTHMA CURRENTLY NOT MANAGED BY AVAILABLE THERAPIES MAY DISPLAY A STRONG INFLAMMATORY COMPONENT DRIVEN BY THE INNATE AND ADAPTIVE IMMUNE SYSTEMS, PARTICULARLY FOLLOWING VIRAL EXACERBATIONS. DEVELOPMENT OF NEW THERAPEUTIC STRATEGIES TARGETING A WIDE RANGE OF INFLAMMATORY MECHANISMS, NOT ONLY TH2 AND TH1/TH17 RESPONSES, IS NEEDED TO CONTROL SEVERE ASTHMA. RAFT PHARMACEUTICALS PROPOSES A NEW TARGET FOR TREATMENT OF SEVERE ASTHMA – OVERABUNDANT AND CLUSTERED, PATHOLOGICAL LIPID RAFTS IN BRONCHIAL EPITHELIAL AND IMMUNE CELLS IN ASTHMATICS. CHOLESTEROL-RICH LIPID RAFTS PROVIDE ORDERED PLASMA MEMBRANE DOMAINS, WHERE RECEPTORS, ION CHANNELS AND ADAPTOR MOLECULES CAN ASSOCIATE TO FORM FUNCTIONAL COMPLEXES. LARGE LIPID RAFTS ARE REQUIRED AS THE LANDING PAD FOR MICROBES ON HOST CELLS AND FOR INITIATION OF MANY INFLAMMATORY PROCESSES IN BRONCHIAL EPITHELIAL AND IMMUNE CELLS. WE DISCOVERED APOA-I BINDING PROTEIN (AIBP) AS A MOLECULE THAT SELECTIVELY TARGETS THE CHOLESTEROL DEPLETION MACHINERY TO PATHOLOGIC LIPID RAFTS IN INFLAMMATORY CELLS, WITHOUT AFFECTING HOMEOSTATIC CELLULAR FUNCTION. THE LATTER EXPLAINS AN EXCEPTIONAL SAFETY PROFILE OF AIBP IN MICE AND RATS. IN THE PHASE I OF THIS GRANT, USING MOUSE MODELS OF ASTHMA, WE DEMONSTRATED THAT ADMINISTRATION OF A BIOLOGIC DERIVED FROM THE AIBP PROTEIN, WAS EFFECTIVE IN AN ACUTE HDM MODEL OF ASTHMA IN MICE, REDUCING AIRWAY HYPERRESPONSIVENESS, AIRWAY EOSINOPHILIA, AND EXPRESSION OF TH2 CYTOKINES AND EPITHELIAL ALARMINS. IN A MODEL OF SEVERE ASTHMA, AIBP SIGNIFICANTLY REDUCED PULMONARY NEUTROPHILIA. FOR PHASE II PROPOSAL, A TEAM OF EXPERTS IN BIOLOGIC DRUG DEVELOPMENT AND IN PRECLINICAL ASSESSMENT AND CLINICAL TRIALS OF THERAPEUTIC AGENTS IN ASTHMA HAS BEEN ASSEMBLED TO FURTHER THE CHARACTERIZATION OF AN IMPROVED AIBP SEQUENCE, RFT1124, AS A DEVELOPMENT CANDIDATE FOR SEVERE ASTHMA. SPECIFICALLY, WE PLAN TO MANUFACTURE AND RELEASE RFT1124 DRUG PRODUCT FOR EFFICACY AND TOXICOLOGY STUDIES USING CGMP-COMPATIBLE PROCESSES AND ANALYTICAL ASSAYS. THE EFFICACY OF RFT1124 AS AN ADD-ON THERAPY TO STANDARD-OF-CARE INHALED CORTICOSTEROIDS AND LONG-ACTING BETA-AGONISTS (ICS+LABA) WILL BE TESTED IN MOUSE MODELS OF ASTHMA EXACERBATED BY RHINOVIRUS (RV) INFECTION. IN ADDITION, WE WILL USE PRECISION CUT LUNG SLICES FROM ASTHMATICS AND NON-ASTHMATICS POSTMORTEM LUNG TISSUE TO TEST THE EFFICACY OF ADD-ON RFT1124 IN ICS+LABA TREATMENT IN RV INFECTED HUMAN AIRWAYS. RNASEQ STUDIES WILL ESTABLISH WHETHER RFT1124 INHIBITS CORTICOSTEROID INSENSITIVE ANTI-INFLAMMATORY PATHWAYS AND/OR ENHANCES CORTICOSTEROID SENSITIVE PATHWAYS IN RV INFECTED HUMAN BRONCHIAL EPITHELIAL CELLS. FURTHER, A NON-GLP SAFETY ASSESSMENT OF INTRANASAL DELIVERY OF RFT1124 WILL BE CONDUCTED TO ESTABLISH AN ESTIMATE OF SAFETY MARGIN AND TARGET ORGANS. THESE RESULTS WILL BE USED TO PREPARE FOR AND CONDUCT PRE-IND (TYPE B) MEETING WITH THE FDA.

THIS TARGETING IS DUE TO AIBP BINDING TO TOLL-LIKE RECEPTOR 4 (TLR4), WHICH IS HIGHLY EXPRESSED ON THE SURFACE OF INFLAMMATORY MICROGLIA, MACROPHAGES AND ACTIVATED DRG NOCICEPTORS. RFT1081-MEDIATED DISRUPTION OF LIPID RAFTS HARBORING ACTIVATED TLR4 ABROGATES THE FACILITATORY CYCLE OF NEUROINFLAMMATION AND NOCICEPTORS’ SPONTANEOUS ACTIVITY AND ALLEVIATES CHRONIC PAIN PHENOTYPES. IN A PRIOR PROJECT, WE DEVELOPED A NON-GMP SCALED-UP UPSTREAM AND DOWNSTREAM MANUFACTURING PROCESS FOR RFT1081 AND CONDUCTED ITS DETAILED CHARACTERIZATION; CONDUCTED PHARMACOKINETICS STUDIES OF SPINALLY DELIVERED RFT1081 IN MICE AND DESIGNED PHARMACODYNAMICS ASSAYS TO EVALUATE RFT1081 TARGET ENGAGEMENT; ESTABLISHED DOSE-DEPENDENT EFFICACY PROFILE FOR AIBP TREATMENT IN CIPN MICE; AND CONDUCTED A NON-GLP DOSE-RANGE TOLERABILITY STUDY OF RFT1081 IN RATS. THESE DATA PROVIDE SUPPORT AND JUSTIFY FURTHER DEVELOPMENT OF RFT1081 IN THE PROPOSED DIRECT-TO-PHASE 2 SBIR STUDIES. IN THIS MILESTONES-DRIVEN PROJECT, WE PLAN TO MANUFACTURE A RFT1081 DRUG PRODUCT LOT FOR TOXICOLOGY STUDIES USING A CGMP-COMPATIBLE PROCESS AND ANALYTICAL ASSAYS. THE RFT1081 DRUG PRODUCT LOT, CONFORMING TO RAFT’S SPECIFICATIONS AND IN OPTIMIZED FORMULATION WILL BE THOROUGHLY CHARACTERIZED FOR STORAGE AND IN-USE STABILITY. THE INITIAL SINGLE-DOSE TOXICOLOGY STUDIES WILL BE PERFORMED IN RATS. WE WILL THEN ALSO EVALUATE THE PHARMACOLOGY, LOCAL AND SYSTEMIC TOXICITY, AND IMMUNE RESPONSE TO REPEATED I.T. ADMINISTRATION OF RAFT1081 IN RATS, DOGS AND NON-HUMAN PRIMATES TO FURTHER STUDY THE SAFETY OF THE DRUG AND TO SELECT A PHARMACOLOGICALLY RELEVANT NON-RODENT SPECIES FOR FURTHER IND- ENABLING PRECLINICAL EVALUATION. RFT1081 WILL ALSO BE EVALUATED IN A MODEL OF CISPLATIN CANCER THERAPY TO ENSURE IT DOES NOT INTERFERE WITH CHEMOTHERAPY ACTION. THE PROJECT WILL CONCLUDE WITH DEVELOPING A DETAILED SYNOPSIS OF AN INTEGRATED FIRST-IN-HUMAN STUDY AND AN OVERALL INDICATION-SUPPORTING CLINICAL STRATEGY, FOLLOWED BY PREPARATION FOR AND CONDUCTING A PRE-IND MEETING WITH THE FDA. WE EXPECT THAT THIS PHASE 2 SBIR PROJECT OUTCOMES WILL INCLUDE: 1) SUCCESSFUL SCALE-UP OF RFT1081 SUITABLE FOR FUTURE CGMP MANUFACTURING, 2) COMPLETION OF DOSE- RANGING TOXICOLOGY STUDIES AND SELECTION OF A RELEVANT NON-RODENT SPECIES, AND 3) A VIABLE IND-ENABLING AND CLINICAL DEVELOPMENT STRATEGY VETTED WITH THE FDA TO DE-RISK FUTURE STEPS OF RFT1081 DEVELOPMENT.

HUMAN TRAFFICKING VICTIMS HAVE COMPLEX LEGAL NEEDS BUT DUE TO A LACK IN IDENTIFICATION, MANY CANNOT ACCESS THE FULL SPECTRUM OF LEGAL RELIEF AVAILABLE TO THEM. EVEN WHEN VICTIMS ARE IDENTIFIED, THE DEMAND FOR LEGAL SERVICES OUTPACES THE NUMBER OF LEGAL PROVIDERS IN THE COUNTRY WHO ARE ADEQUATELY TRAINED IN TRAUMA-INFORMED AND RACIALLY EQUITABLE SERVICES. AS A RESULT, LEGAL SERVICES ARE AN UNMET NEED OF TRAFFICKING VICTIMS AND COMPRESSED TRAINING ON THE LEGAL NEEDS OF AND ETHICAL PROVISION SERVICES TO TRAFFICKING VICTIMS IS IN HIGH DEMAND. THROUGH ITS EXPERT LEGAL AND SOCIAL SERVICES STAFF, THE COALITION TO ABOLISH SLAVERY AND TRAFFICKING (CAST) PROPOSES TO CONTINUE ITS COMPREHENSIVE, NATIONAL, ACCESSIBLE AND INCLUSIVE TRAINING AND TECHNICAL ASSISTANCE (TTA) PROJECT TO STRENGTHEN LEGAL SERVICE PROVIDERS, PRO BONO ATTORNEYS, AND SOCIAL SERVICE PROVIDERS CAPACITY TO IDENTIFY AND PROVIDE COMPREHENSIVE LEGAL SERVICES TO VICTIMS OF HUMAN TRAFFICKING THROUGH A TRAUMA-INFORMED AND RACE EQUITY LENS. IN PARTNERSHIP WITH THE OFFICE FOR VICTIMS OF CRIME, CAST WILL BUILD ON OVER 15 YEARS OF PROVIDING TTA TO DEEPEN AND EXPAND ITS NATIONAL TTA EFFORTS TO INCREASE TRAFFICKING VICTIMS ACCESS TO QUALITY COMPREHENSIVE LEGAL SERVICES. ADDITIONALLY, CAST WILL LEVERAGE COLLABORATION WITH MANY NATIONAL PARTNERS LIKE FREEDOM NETWORK USAS SURVIVOR REENTRY PROJECT TO PROVIDE COMPREHENSIVE TTA. THE PROJECT WILL PROVIDE COMPREHENSIVE LIVE AND ON-DEMAND TRAINING TO LEGAL AND SOCIAL SERVICE PROVIDERS INCLUDING ANTI-TRAFFICKING LAWYERING TRAINING MODULE 2, INTENSIVE T VISA DECLARATION WORKSHOPS, AND VICTIMS RIGHTS ADVOCACY FOR SOCIAL SERVICE PROVIDERS, AS WELL AS SELF-PACED ONLINE TRAINING TO MANY MORE ATTORNEYS AND SOCIAL SERVICE PROVIDERS NATIONALLY THROUGH AN ADVANCED LEARNING MANAGEMENT SYSTEM FOR ASYNCHRONOUS AND SYNCHRONOUS LEARNING. CAST WILL ALSO CONTINUE TO PROVIDE INDIVIDUAL TECHNICAL ASSISTANCE TO 40 LEGAL AND SOCIAL SERVICE PROFESSIONALS A MONTH, AS WELL AS WEEKLY LEGAL WORKING GROUP CALLS AND MONTHLY OFFICE HOURS FOR SOCIAL SERVICE PROVIDERS. CAST WILL SUBGRANT TO FREEDOM NETWORK USA TO SUPPORT TTA ON CRIMINAL RECORD RELIEF, AND TO THE NATIONAL CRIME VICTIMS LAW INSTITUTE WHO WILL SUPPORTTTA ON CRIME VICTIMS RIGHTS. WHILE TARGETED BENEFICIARIES ARE LEGAL AND SOCIAL SERVICE PROVIDERS, ANYONE INTERESTED IN INCREASING KNOWLEDGE IN THIS AREA WILL BE ABLE TO ACCESS THE CAST TTA WEBSITE FOR MATERIALS ON POPULAR TOPICS. THROUGH THIS PROJECT, CAST WILL HAVE THE OPPORTUNITY TO CONTINUE TO TRANSFORM THE LANDSCAPE OF TRAINED LEGAL SERVICE PROVIDERS ASSISTING VICTIMS OF HUMAN TRAFFICKING AND TO VASTLY IMPROVE VICTIMS LIVES.

METABOQUEST: A SUITE OF TOOLS FOR METABOLITE ANNOTATION - METABOQUEST: A SUITE OF TOOLS FOR METABOLITE ANNOTATION PROJECT SUMMARY METABOLOMICS AIMS AT HIGH THROUGHPUT DETECTION, QUANTIFICATION, AND IDENTIFICATION OF METABOLITES IN BIOLOGICAL SAMPLES. THE USE OF LIQUID CHROMATOGRAPHY COUPLED WITH MASS SPECTROMETRY (LC-MS) HAS RISEN IN PROMINENCE IN THE FIELD OF METABOLOMICS DUE TO ITS ABILITY TO ANALYZE A SIZABLE NUMBER OF METABOLITES WITH A LIMITED AMOUNT OF BIOLOGICAL MATERIAL. HOWEVER, IN A TYPICAL UNTARGETED METABOLOMICS ANALYSIS OF HUMAN SAMPLES BY LC-MS, ABOUT 70% OF THE DETECTED PEAKS REPRESENT UNKNOWN ANALYTES MAINLY BECAUSE EXISTING MASS SPECTRAL LIBRARIES COVER ONLY A SMALL FRACTION OF KNOWN COMPOUNDS, BUT ALSO DUE TO UNCERTAINTY IN PEAK PICKING, ALIGNMENT OF PEAKS, AND RECOGNIZING ISOTOPIC PEAKS AND ADDUCT FORMS. THESE CHALLENGES HAVE KEPT AT BAY THE PACE OF DEVELOPMENT OF DATA ANALYTICS PIPELINES FOR METABOLOMICS AND ITS INTEGRATION WITH OTHER OMICS STUDIES. THE GOAL OF THIS PHASE II SBIR PROPOSAL IS TO MAKE METABOLOMICS STUDIES ON A PAR WITH OTHER OMICS STUDIES SUCH AS GENOMICS, TRANSCRIPTOMICS, AND PROTEOMICS, FOR WHICH WELL-ESTABLISHED PIPELINES ARE AVAILABLE. BY DOING SO, WE WILL ACCELERATE THE ROLE OF METABOLOMICS IN SYSTEMS BIOLOGY APPROACHES FOR VARIOUS APPLICATIONS INCLUDING BIOMARKER AND DRUG DISCOVERY. TO ACHIEVE THIS GOAL, WE PROPOSE TO DEVELOP A CLOUD-BASED PLATFORM THAT ALLOWS CUSTOMERS TO BUILD PIPELINES FOR ANALYSIS OF LC-MS-BASED UNTARGETED METABOLOMICS DATA, STARTING FROM PEAK DETECTION TO METABOLITE ANNOTATION. THIS WILL BE ACCOMPLISHED BY IMPLEMENTING A SUITE OF INNOVATIVE TOOLS THAT CAN BE ASSEMBLED INTO CUSTOMIZED PIPELINES AND BY ENHANCING METABOLITE ANNOTATION ACCURACY THROUGH INTEGRATION OF INFORMATION DERIVED FROM MULTIPLE RESOURCES INCLUDING COMPOUND DATABASES, PATHWAYS, BIOCHEMICAL NETWORKS, AND MASS SPECTRAL LIBRARIES. AIM 1 OF THIS PROPOSAL WILL FOCUS ON DEVELOPING A SUITE OF TOOLS TO ENABLE: (1) PEAK DETECTION, ALIGNMENT, AND QUALITY ASSESSMENT; (2) ADDUCT AND ISOTOPIC PEAK RECOGNITION; (3) MASS-BASED SEARCH AGAINST MULTIPLE COMPOUND DATABASES; (4) EXPERT-BASED EVALUATION OF PUTATIVE IDS; (5) ISOTOPIC PATTERN ANALYSIS; (6) NETWORK-BASED EVALUATION OF PUTATIVE IDS; (7) SPECTRAL MATCHING OF MS/MS DATA AGAINST EXPERIMENTAL AND IN- SILICO FRAGMENTATION PATTERNS; (8) DEEP LEARNING-BASED PREDICTION OF COMPOUND FINGERPRINTS; AND (9) INTEGRATIVE ASSESSMENT OF PUTATIVE METABOLITE IDS VIA A PROBABILISTIC MODEL. AIM 2 WILL ASSEMBLE THE TOOLS DEVELOPED IN AIM 1 INTO A CLOUD-BASED PLATFORM, METABOQUEST, WHICH PROVIDES USERS WITH INTERACTIVE VISUALIZATION OF PEAKS, ISOTOPIC PATTERNS, NETWORKS, AND MASS SPECTRA. FURTHERMORE, AIM 2 WILL FOCUS ON INTEGRATING INTO METABOQUEST A PIPELINE BUILDER THAT ALLOWS USERS TO CREATE PIPELINES BY LINKING MODULES AND RUN THEM REMOTELY THROUGH A MODULAR INTERACTIVE WEB INTERFACE. AIM 3 WILL PERFORM A COMPREHENSIVE EVALUATION OF METABOQUEST IN TERMS OF METABOLITE ANNOTATION ACCURACY, NUMBER OF ANNOTATED METABOLITES, AND COMPUTATIONAL EFFICIENCY COMPARED TO OTHER EXISTING TOOLS. ACCURACY IN METABOLITE ANNOTATION WILL BE EVALUATED VIA EXPERIMENTAL METHODS IN WHICH MS/MS DATA FROM UNKNOWN ANALYTES AND REFERENCE COMPOUNDS ARE COMPARED, AND BY USING LC-MS/MS DATA FROM MULTIPLE METABOLOMICS STUDIES THAT CONSIST OF GROUND-TRUTH INFORMATION. SUCCESSFUL IMPLEMENTATION AND VALIDATION OF METABOQUEST WILL CONTRIBUTE TO ADDRESSING THE MAJOR BOTTLENECK IN METABOLOMICS - METABOLITE IDENTIFICATION, THEREBY ELIMINATING THE NEED FOR MANUAL VERIFICATION OF PUTATIVE METABOLITE IDS AND ENHANCING THE CONTRIBUTION OF METABOLOMICS STUDIES, SPECIFICALLY IN DISEASE BIOMARKER AND DRUG DISCOVERY.
