THIS ADDRESSES THE FUNDING PREFERENCE OF THIS GRANT, UNDER QUALIFICATION 2, FOCUSED ON SUBSTANTIALLY BENEFITING UNDERSERVED POPULATIONS. THE POPULATION OF CLINICAL FACULTY AND PRECEPTORS THAT WILL BE RECRUITED AND TRAINED THROUGH THIS PROPOSED PROJECT INCLUDE FEDERALLY DESIGNATED MEDICALLY UNDERSERVED AREAS (MUA) AND MEDICALLY UNDERSERVED POPULATIONS (MUP) THROUGHOUT ILLINOIS AND THE HHS REGION 5 STATES, AS NOTED IN OUR FUNDING PREFERENCE REQUEST. THE OBJECTIVES OF THIS PROJECT ARE: 1) ESTABLISH A SUSTAINABLE CLINICAL FACULTY AND PRECEPTOR TRAINING PROGRAM TO EXPAND THE NURSING WORKFORCE SERVING RURAL AND UNDERSERVED POPULATIONS IN ILLINOIS AND THE HHS REGION 5 STATES; 2) FOSTER A LEARNING EXPERIENCE THAT PROMOTES KNOWLEDGE BUILDING AND ENHANCEMENT OF CLINICAL FACULTY AND PRECEPTOR ABILITIES, RESILIENCY, AND EMOTIONAL INTELLIGENCE; 3) MAINTAIN, CREATE, AND EXPAND COLLABORATIVE PARTNERSHIPS BETWEEN ACADEMIA AND PRACTICE TO PROMOTE CLINICAL FACULTY AND PRECEPTOR EDUCATION TO INCREASE THE NUMBER OF CLINICAL FACULTY AND PRECEPTORS TRAINING NURSES IN RURAL AND UNDERSERVED COMMUNITIES; 4) INCREASE THE RECRUITMENT OF CLINICAL FACULTY AND PRECEPTORS WORKING IN RURAL AND UNDERSERVED COMMUNITIES WITHIN HHS REGION 5. GRADUATE NURSING STUDENTS ENROLLED IN NURSE EDUCATOR PROGRAMS, NURSE PRACTITIONER PROGRAMS, AND NURSE ANESTHESIA PROGRAMS, CLINICAL FACULTY, REGISTERED NURSES AND ADVANCED PRACTICE NURSES WORKING IN INPATIENT AND OUTPATIENT SETTINGS, AND NEW AND EXISTING PRECEPTORS IN HHS REGION 5 STATES WILL BE THE TARGETED POPULATION FOR THIS PROJECT. STUDENTS ENROLLED IN GRADUATE NURSING PROGRAMS ARE OFTEN EXPERIENCED REGISTERED NURSES WHO ARE ALREADY OR WILL SOON TAKE ON PRECEPTOR ROLES AND CLINICAL FACULTY ROLES IN THE NURSING WORKFORCE. CUR RENT PRECEPTORS IN THE COMMUNITY AND CLINICAL FACULTY ARE OFTEN ENGAGED IN THE ROLE OF PRECEPTOR WITHOUT FORMAL TRAINING AND LACK SUPPORT. THE PROJECT SEEKS TO REACH POPULATIONS THROUGH THE DEVELOPMENT OF EASILY ACCESSIBLE SELF-PACED INTERNET-BASED TRAINING MODULES, AN ANNUAL CLINICAL FACULTY AND PRECEPTOR CONFERENCE, AND INCORPORATION OF EMOTIONAL INTELLIGENCE AND RESILIENCY COACHING. THESE MODULES WILL COVER TOPICS IMPORTANT FOR CLINICAL FACULTY AND PRECEPTORS INCLUDING METHODS OF GIVING CONSTRUCTIVE FEEDBACK, UNCONSCIOUS BIAS, CHALLENGING STUDENT SITUATIONS, BUILDING STUDENT INDEPENDENCE AND CONFIDENCE, EMOTIONAL INTELLIGENCE, RESILIENCY, AND DEVELOPING CLINICAL STUDENT LEARNING AND EVALUATION PLANS. THE CLINICAL FACULTY AND PRECEPTOR ANNUAL CONFERENCE WILL BE HOSTED AT THE SIUE CAMPUS USING A HYBRID FORMAT TO FACILITATE ATTENDANCE FOR ALL PARTICIPANTS WITHIN HHS REGION 5. EACH YEAR THE PROGRAM’S GOAL IS TO TRAIN 60 STUDENTS IN GRADUATE NURSE EDUCATOR, NURSE ANESTHESIA, OR NURSE PRACTITIONER PROGRAMS, 100 HEALTHCARE PRECEPTORS AND 40 CLINICAL FACULTY FOR EACH YEAR OF THE PROJECT PROGRAM FUNDING. WHILE IDENTIFYING THE NEURAL UNDERPINNINGS OF ASD HAS BEEN A MAJOR FOCUS IN THE FIELD OF GENETICS AND NEUROSCIENCE, IT IS NOT CLEAR WHICH BRAIN REGION (S) AND CELL TYPES MAY BE FUNCTIONING ABNORMALLY TO GIVE RISE TO ASD. THE CEREBELLUM, A BRAIN REGION TYPICALLY CONSIDERED TO BE INVOLVED IN MOTOR COORDINATION AND CONTROL, HAS RECEIVED CONSIDERABLE ATTENTION FOR ITS POTENTIAL ROLE IN ASD, WITH A GROWING BODY OF CLINICAL EVIDENCE CORRELATING ASD DIAGNOSES WITH ABNORMALITIES IN CEREBELLAR ANATOMY, FUNCTION, AND MOTOR OR VESTIBULAR BEHAVIORS/FUNCTIONS. IN PARALLEL, CEREBELLAR INVOLVEMENT IN CIRCUITS OUTSIDE OF THE MOTOR SYSTEM PROVIDES A SUBSTRATE FOR THE CEREBELLUM TO INFLUENCE NON-MOTOR BEHAVIORS AND PROCESSES, SETTING THE STAGE FOR THE IMPORTANCE OF APPROPRIATE CEREBELLAR FUNCTION IN A HOST OF NEURAL PROCESSES. IT IS ALSO NOT CLEAR WHAT ASPECTS OF CEREBELLAR GRANULE CELL FUNCTION AND CONNECTIVITY MAY BE IMPORTANT FOR SHAPING CERTAIN NON-MOTOR (AND MOTOR) BEHAVIORS. TO ADDRESS THIS KNOWLEDGE GAP, IN AIMS 1 AND 2 WE PROPOSE TO IDENTIFY THE DEGREE TO WHICH AN ASD-LINKED GENE DETERMINES PROPERTIES OF CEREBELLAR GRANULE CELL SYNAPTIC INTERACTIONS AND CORTICAL CIRCUITRY DYNAMICS OVER TIME. SIUC IS A RURAL UNIVERSITY AND THE MAJOR RESEARCH INSTITUTION IN SOUTHERN ILLINOIS, ONE OF AMERICA?S MOST ECONOMICALLY DISTRESSED REGIONS. OVER ITS FIVE-YEAR DURATION, THIS PROJECT WILL FUND SCHOLARSHIPS TO 40 UNIQUE FULL-TIME TRANSFER STUDENTS WHO ARE PURSUING BACHELOR?S DEGREES IN GEOGRAPHY & ENVIRONMENTAL RESOURCES AND GEOLOGY. TRANSFER STUDENTS WILL RECEIVE TWO-YEAR SCHOLARSHIPS. THE PROPOSED PROJECT IS AN INTERDISCIPLINARY STEM PROGRAM, EARTH-SUSTAINABILITY SCHOLARSHIP (ESS), FOR ACADEMICALLY TALENTED STUDENTS WHO DEMONSTRATE FINANCIAL NEED TO PURSUE UNDERGRADUATE ENVIRONMENTAL AND EARTH SCIENCE DEGREES. A SUSTAINABILITY-MINDED STEM WORKFORCE IS VITAL FOR THE NATION TO STRATEGICALLY DEVELOP NATURAL RESOURCES, EXPERIENCE ECONOMIC GROWTH, AND MAKE INFORMED DECISIONS IN A RAPIDLY CHANGING WORLD. THE PROJECT WILL DEVELOP AN EDUCATIONAL PIPELINE FROM STUDENT ADMISSION AND TRAINING TO GRADUATION AND PROFESSIONAL DEVELOPMENT AND ADVANCE THE UNDERSTANDING OF THE DOMINANT FACTORS THAT INCREASE THE ACADEMIC AND PROFESSIONAL SUCCESS OF LOW-INCOME STUDENTS. A MENTORING SYSTEM AND A NETWORKED SUPPORTING COMMUNITY THAT INTEGRATE CAMPUS RESOURCES AND SUSTAINABILITY ACTIVITIES WILL BE ESTABLISHED FOR IMPROVED CURRICULAR AND CO-CURRICULAR EXPERIENCES. THE PROJECT IS EXPECTED TO BROADEN THE PARTICIPATION OF LOW-INCOME STUDENTS IN ENVIRONMENTAL AND EARTH SCIENCE BY REDUCING FINANCIAL PRESSURE, INCREASING LEARNING OPPORTUNITIES AND OUTCOMES, AND BRIDGING OVERALL CAREER SUCCESS. IT WILL ALSO ADVANCE THE MISSION OF U.S. INSTITUTIONS OF HIGHER EDUCATION THAT TRAIN A DIVERSE AND GLOBALLY COMPETITIVE STEM WORKFORCE IN SUSTAINABLE ENVIRONMENTAL AND EARTH SCIENCE. THE OVERALL GOAL OF THIS PROJECT IS TO INCREASE STEM DEGREE COMPLETION OF LOW-INCOME, HIGH-ACHIEVING UNDERGRADUATES WITH DEMONSTRATED FINANCIAL NEED. SPECIFIC OBJECTIVES INCLUDE: (1) INCREASING EDUCATIONAL OPPORTUNITIES FOR ACADEMICALLY TALENTED STUDENTS WITH FINANCIAL NEED, (2) IMPROVING EDUCATIONAL SUPPORT PROGRAMS AT SIUC, (3) IMPROVING THE RETENTION OF STUDENTS IN THE SIUC SCHOOL OF EARTH SYSTEMS AND SUSTAINABILITY, AND (4) ENHANCING THE FUTURE STEM WORKFORCE WITH SKILLS IN EARTH SCIENCE AND SUSTAINABILITY. TO ADDRESS THE HISTORIC DISCONNECT BETWEEN SUSTAINABILITY AND EARTH SCIENCE TRAINING, IT IS VITAL TO INTEGRATE SUSTAINABILITY INTO EARTH SCIENCE EDUCATION IN INSTITUTES OF HIGHER EDUCATION. ADVANCING UNDERSTANDING OF HOW SUPPORTING SERVICES AND PROGRAMS DRIVE ACADEMIC SUCCESS IN GEOSCIENCE MAJORS, ESPECIALLY FOR TRANSFER STUDENTS, IS ALSO CRITICAL. THIS PROJECT WILL ESTABLISH AN EDUCATIONAL PROTOTYPE THAT SUCCESSFULLY SUPPORTS ACADEMICALLY TALENTED AND LOW-INCOME TRANSFER STUDENTS TO BECOME SUSTAINABILITY-FOCUSED EARTH SCIENCE DEGREE GRADUATES. FOLLOWING THE THEORY OF PLANNED BEHAVIORS, THE SUPPORTING SERVICE AND ACTIVITIES TO ENHANCE ATTITUDES, SUBJECTIVE NORMS, AND PERCEIVED BEHAVIORAL CONTROL WILL BE FRAMED AS THREE CRITICAL CONSTRUCTS THAT ARE EXPECTED TO DRIVE STUDENTS? POSITIVE INTENTIONS FOR GRADUATION. THE PROJECT HOPES TO CREATE AN INCLUSIVE AND DIVERSE LEARNING COMMUNITY AND INFRASTRUCTURE TO ADVANCE THE PROJECT GOALS AND MISSIONS. IT IS EXPECTED THAT 40 LOW-INCOME ACADEMICALLY TALENTED TRANSFER STUDENTS WILL BE AWARDED $10,000 SCHOLARSHIPS EACH YEAR AND THAT AN OVERALL 85% RETENTION RATE AND 80% GRADUATION RATE WILL BE ACHIEVED. THE PROJECT WILL UTILIZE A MANAGEMENT-ORIENTED FORMATIVE AND SUMMATIVE EVALUATION APPROACH. RESULTS WILL BE DISSEMINATED INTERNALLY TO THE OFFICE OF THE PROVOST AND FACULTY MEMBERS AT SIUC AND SHARED EXTERNALLY TO THE STEM COMMUNITY THROUGH STEM CENTRAL, SOCIAL MEDIA, PROFESSIONAL CONFERENCES, AND PUBLICATIONS IN STEM EDUCATION JOURNALS. THIS PROJECT IS FUNDED BY NSF?S SCHOLARSHIPS IN SCIENCE, TECHNOLOGY, ENGINEERING, AND MATHEMATICS PROGRAM, WHICH SEEKS TO INCREASE THE NUMBER OF LOW-INCOME ACADEMICALLY TALENTED STUDENTS WITH DEMONSTRATED FINANCIAL NEED WHO EARN DEGREES IN STEM FIELDS. LOUIS COMMUNITY COLLEGE, CITY COLLEGES OF CHICAGO-MALCOLM X COLLEGE, OAKTON COLLEGE, AND SHAWNEE COMMUNITY COLLEGE INTEND TO CONDUCT A WIDE-RANGE OF PROFESSIONAL DEVELOPMENT ACTIVITIES. THEY ALSO AIM TEST A MODEL OF PROFESSIONAL DEVELOPMENT, AND DETERMINE THE EFFECTS OF INSTITUTIONAL, DISCIPLINARY, INSTRUCTOR, AND STUDENT CHARACTERISTICS ON THE EFFECTIVENESS OF FLIPPED INSTRUCTION IN STEM UNDERGRADUATE EDUCATION. FOUR PAIRINGS OF INSTITUTIONS - EACH PAIR INCLUDING A 4-YEAR INSTITUTION AND A 2-YEAR INSTITUTION - CONSTITUTE THE FOUNDATION FOR BOTH IMPLEMENTATION AND INVESTIGATION. FLIPPED INSTRUCTION HAS DEMONSTRATED ITS POTENTIAL TO SIGNIFICANTLY TRANSFORM TEACHING AND LEARNING IN UNDERGRADUATE STEM EDUCATION. DRAWING ON EXTENSIVE RESEARCH AND FINDINGS FROM A PRIOR NSF IUSE EXPLORATORY AWARD, THIS TRACK 2 INSTITUTIONAL AND COMMUNITY TRANSFORMATION LEVEL II COLLABORATIVE RESEARCH PROJECT INTENDS TO (1) MONITOR STUDENT- CENTERED ORIENTATION OF FACULTY, IMPLEMENTATION, AND VARIATIONS IN IMPLEMENTATION BASED ON INSTITUTIONAL TYPE, DEPARTMENT/DISCIPLINE, AND STUDENT DEMOGRAPHICS; (2) IDENTIFY ENABLERS AND OBSTACLES; AND (3) EXAMINE CHANGE LEADERSHIP AS FOUNDATIONAL STRATEGIES FOR DEVELOPING A COMMUNITY OF PRACTICE. ACTIVITIES HAVE BEEN DESIGNED TO ENHANCE INSTRUCTOR EXPERTISE AND SKILLS IN FLIPPED INSTRUCTION TO MEET THE NEEDS OF STUDENTS AND LEADERSHIP TO DRIVE TRANSFORMATIVE, LASTING CHANGE WITHIN THE RESPECTIVE INSTITUTIONS. GROUNDED IN A COMMUNITIES OF TRANSFORMATION PERSPECTIVE, THE MIXED METHODS RESEARCH DESIGN USES RIGOROUS DATA COLLECTION, ANALYSIS, AND INTERPRETATION METHODS. THE TEAM WILL DOCUMENT AND ASSESS INSTRUCTORS' CONFIDENCE, ATTITUDES, AND MOTIVATIONS TO USE FLIPPED INSTRUCTION, AS WELL AS STUDENTS' ACADEMIC PERFORMANCE AND PERSISTENCE IN STEM. HOWEVER, EVEN WITH THE BEST CHEMICALLY-DEFINED DIFFERENTIATION PROTOCOL OF PLURIPOTENT STEM CELLS, THE CONTROL OF CELL-LINEAGE SPECIFICATION REMAINS POOR. BESIDES CHEMICAL SIGNALING, IT IS NOW WIDELY ACCEPTED THAT PHYSICAL SIGNALS FROM THE EXTRACELLULAR MATRIX (ECM) PLAY A CRUCIAL ROLE IN CELL FATE DETERMINATION. NEVERTHELESS, CONTROL OF CELL-LINEAGE SPECIFICATION BY SUCH MECHANICAL FORCES ALONE COULD NOT BE IMPROVED POSSIBLY DUE TO THE LACK OF PRECISE CONTROL OF FORCES AT THE SINGLE-MOLECULE LEVEL AND THE LACK OF SYNERGY BETWEEN CHEMICAL SIGNALING AND MECHANICAL PATHWAYS. TO ADDRESS THIS GAP, THE PROPOSED STUDY AIMS TO PROVIDE A MECHANISTIC FRAMEWORK OF SINGLE EPISC FATE DECISIONS (SELF-RENEWAL AND DIFFERENTIATION) BASED ON CHEMICAL AND SINGLE-MOLECULE FORCE BASED APPROACHES. THE CENTRAL HYPOTHESIS IS THAT THE SYNERGISTIC EFFECT OF CHEMICAL AND SINGLE-MOLECULE FORCE CUES VIA CELL-ECM AND CELL-CELL INTERACTIONS CAN CONTROL FATE DECISIONS FAR MORE EFFECTIVELY THAN PREVIOUSLY POSSIBLE. THE LONG-TERM GOAL IS TO DEVELOP NOVEL APPROACHES TO CONTROL THE DIRECTED DIFFERENTIATION OF PLURIPOTENT CELLS INTO ALL THREE GERM-LAYERS. TO THIS END, THE FOLLOWING THREE AIMS ARE PROPOSED. SPECIFICALLY, AIM 1 WILL FOCUS ON UNDERSTANDING THE MECHANISM OF SINGLE-MOLECULE FORCE MEDIATED DIFFERENTIATION OF EPISCS INTO THE MESODERM LINEAGE. THE FORCE TRANSMISSION INTO SINGLE CELLS VIA SINGLE AVSS3 INTEGRINS WILL BE CONTROLLED BY TENSION GAUGE TETHERS. THESE DNA-BASED RUPTURABLE TETHERS CAN PRECISELY LIMIT THE AMOUNT OF FORCE AT THE SINGLE-MOLECULE LEVEL. TOGETHER WITH CHEMICAL SIGNALING, SUCH PRECISE CONTROL AND SPECIFIC TARGETING OF MECHANICAL PATHWAYS MAY LEAD TO SUPERIOR CONTROL OF CELL DIFFERENTIATION INTO THE MESODERM. IN AIM 2, THE MECHANISM OF SELF-RENEWAL OF SINGLE EPISCS WILL BE IDENTIFIED BY DEFINING A MICROENVIRONMENT COMPOSED OF SELF-RENEWAL PROMOTING LIGANDS SUCH AS E-CADHERIN. IN AIM 3, DIFFERENTIATION OF SINGLE EPISCS WILL BE DEFINED VIA THE NOTCH PATHWAY BY ENGINEERED LOW- TOLERANCE TENSION GAUGE TETHER CALLED “NANO-YOYO” TO ACTIVATE FORCE-DEPENDENT NOTCH SIGNALING. ADDITIONALLY, THERE IS A NEED FOR INCREASED KNOWLEDGE IN LONG-TERM CARE, GERIATRIC SUPPORT, AND COMMUNITY NURSING TO BETTER SERVE RURAL AND ADJACENT POPULATIONS. THIS PROPOSED PROJECT HAS TWO SPECIFIC AIMS WITH SEVERAL OBJECTIVES TO ACHIEVE THESE AIMS SUCCESSFULLY. AIM 1: EXPAND NURSING WORKFORCE CAPACITY IN RURAL AND LONG-TERM CARE SETTINGS. OBJECTIVES TO ADDRESS AIM 1 INCLUDE: 1) COLLABORATE WITH COMMUNITY COLLEGES, UNIVERSITIES, AND CLINICAL PARTNERS IN RURAL AREAS FOR RECRUITMENT OUTREACH PROGRAMS IN MIDDLE AND HIGH SCHOOLS TO PROMOTE MULTIPLE PATHWAYS TO NURSING DEGREE COMPLETION. 2) IMPLEMENT INTENTIONAL RECRUITMENT EFFORTS FOCUSING ON DIVERSIFYING THE NURSING WORKFORCE. 3) PROVIDE FUNDING OPPORTUNITIES FOR STUDENTS IN THE ACCELERATED POST-BACCALAUREATE NURSING PROGRAM. ONE SUCH PREDISPOSING FACTOR IS BEHAVIORAL SENSITIVITY TO THE AVERSIVE ATTRIBUTES OF COCAINE, WHICH OVERLAP WITH THE REWARDING EFFECTS TO PROTECT AGAINST COCAINE-SEEKING. PRELIMINARY DATA IN THIS PROPOSAL SUGGEST THAT THE AVERSIVE EFFECT OF COCAINE MAY BE MUCH STRONGER IN SOME INDIVIDUALS THAN OTHERS AND POTENTIALLY A DETERMINANT FOR COCAINE ADDICTION VULNERABILITY. THE UNDERLYING CELLULAR MECHANISM OF THIS VARIABILITY REMAINS UNKNOWN. HOWEVER, OUR PRELIMINARY DATA REVEALED THAT HIGHER RHOA ACTIVITY AND D2-MSN EXCITABILITY IN THE MNACSHELL CORRELATE WITH RATS DISPLAYING HIGHER AVERSIVE EFFECTS OF COCAINE (AND CONSEQUENTLY LOWER DRIVE TO SEEK COCAINE). NEVERTHELESS, IT IS UNKNOWN IF THESE CELLULAR AND MOLECULAR ADAPTATIONS IN THE MNACSHELL ARE INVOLVED IN THE AVERSIVE ATTRIBUTES OF COCAINE. ACCORDINGLY, TO ADDRESS THIS GAP IN KNOWLEDGE, I’M PROPOSING TWO AIMS. IN AIM 1, I WILL DETERMINE THE ROLES OF D2MSN ON COCAINE AVOIDANCE BEHAVIOR, AND FOR THIS, I WILL BE TRAINED IN FIBER PHOTOMETRIC AND OPTOGENETIC METHODS. I HYPOTHESIZE THAT D2- BUT NOT D1-MSNS ARE ACTIVATED DURING COCAINE’S AVERSIVE PHASE, AND THAT D2 ACTIVITY IN TURN DRIVES CONDITIONED NEGATIVE ATTRIBUTES OF COCAINE. IN AIM 2, I WILL EXAMINE THE ROLES OF RHOA AND RELATED GENES ON NEURAL EXCITABILITY AND COCAINE AVOIDANCE IN A CELL-SPECIFIC MANNER. FIRST, I WILL USE MRNA TRAP METHODS TO ANALYZE CELL-SPECIFIC GENE TRANSLATION ASSOCIATED WITH RHOA SIGNALING OR EXCITABILITY LINKED TO COCAINE AVOIDANCE PHENOTYPES. IN ADDITION, TO TEST THE CAUSAL ROLE OF RHOA IN EXCITABILITY AND COCAINE AVOIDANCE BEHAVIOR, I WILL DECREASE OR INCREASE RHOA ACTIVITY IN A CELL-SPECIFIC MANNER USING AN ADENO-ASSOCIATED GENE MANIPULATION STRATEGY. I HYPOTHESIZE THAT INCREASES IN RHOA ACTIVITY IN D2-MSN BUT NOT D1-MSN ENHANCES EXCITABILITY IN THE MNACSHELL CONFER PROTECTION AGAINST THE ACQUISITION OF COCAINE-SEEKING. EVIDENCE SHOWS THAT CLINICAL, CLASSROOM AND SIMULATION EXPERIENCES THAT EMPHASIZE CONCEPTS FROM SOCIAL DETERMINANTS OF HEALTH PREPARE UNDERGRADUATE NURSES FOR APPROPRIATE AND INCLUSIVE CARE OF DIVERSE POPULATIONS. IN ADDITION, LITERATURE IS ABUNDANT WITH STRATEGIES TO RECRUIT AND RETAIN NURSES OF DIVERSE BACKGROUNDS WHICH INCLUDES MENTORING, ACADEMIC, SOCIAL AND PERSONAL ADVISING, AND SUPPORTIVE ACADEMIC ENVIRONMENTS. OBJECTIVES OF THIS PROJECT INCLUDE: 1) RECRUITMENT OF DIVERSE NURSING STUDENTS THROUGH DEVELOPMENT OF A SCHOOL PIPELINE IN COLLABORATION WITH SCHOOL DISTRICTS AND LOCAL HOSPITALS TO FACILITATE AWARENESS OF THE NURSING PROFESSION BY OFFERING SHADOWING EXPERIENCES AND MOBILE SIMULATION EXPERIENCES. NURSE EDUCATION PRACTICE QUALITY AND RETENTION: CLINICAL FACULTY AND PRECEPTOR ACADEMY - THE PRIMARY GOAL OF THIS PROJECT IS TO RECRUIT AND TRAIN CLINICAL FACULTY AND PRECEPTORS IN LOCAL AND REGIONAL COMMUNITY HEALTHCARE AND ACADEMIC ORGANIZATIONS WHICH SERVE RURAL AND MEDICALLY UNDERSERVED POPULATIONS. THIS ADDRESSES THE FUNDING PREFERENCE OF THIS GRANT, UNDER QUALIFICATION 2, FOCUSED ON SUBSTANTIALLY BENEFITING UNDERSERVED POPULATIONS. THE POPULATION OF CLINICAL FACULTY AND PRECEPTORS THAT WILL BE RECRUITED AND TRAINED THROUGH THIS PROPOSED PROJECT INCLUDE FEDERALLY DESIGNATED MEDICALLY UNDERSERVED AREAS (MUA) AND MEDICALLY UNDERSERVED POPULATIONS (MUP) THROUGHOUT ILLINOIS AND THE HHS REGION 5 STATES, AS NOTED IN OUR FUNDING PREFERENCE REQUEST. THE OBJECTIVES OF THIS PROJECT ARE: 1) ESTABLISH A SUSTAINABLE CLINICAL FACULTY AND PRECEPTOR TRAINING PROGRAM TO EXPAND THE NURSING WORKFORCE SERVING RURAL AND UNDERSERVED POPULATIONS IN ILLINOIS AND THE HHS REGION 5 STATES; 2) FOSTER A LEARNING EXPERIENCE THAT PROMOTES KNOWLEDGE BUILDING AND ENHANCEMENT OF CLINICAL FACULTY AND PRECEPTOR ABILITIES, RESILIENCY, AND EMOTIONAL INTELLIGENCE; 3) MAINTAIN, CREATE, AND EXPAND COLLABORATIVE PARTNERSHIPS BETWEEN ACADEMIA AND PRACTICE TO PROMOTE CLINICAL FACULTY AND PRECEPTOR EDUCATION TO INCREASE THE NUMBER OF CLINICAL FACULTY AND PRECEPTORS TRAINING NURSES IN RURAL AND UNDERSERVED COMMUNITIES; 4) INCREASE THE RECRUITMENT OF CLINICAL FACULTY AND PRECEPTORS WORKING IN RURAL AND UNDERSERVED COMMUNITIES WITHIN HHS REGION 5. GRADUATE NURSING STUDENTS ENROLLED IN NURSE EDUCATOR PROGRAMS, NURSE PRACTITIONER PROGRAMS, AND NURSE ANESTHESIA PROGRAMS, CLINICAL FACULTY, REGISTERED NURSES AND ADVANCED PRACTICE NURSES WORKING IN INPATIENT AND OUTPATIENT SETTINGS, AND NEW AND EXISTING PRECEPTORS IN HHS REGION 5 STATES WILL BE THE TARGETED POPULATION FOR THIS PROJECT. STUDENTS ENROLLED IN GRADUATE NURSING PROGRAMS ARE OFTEN EXPERIENCED REGISTERED NURSES WHO ARE ALREADY OR WILL SOON TAKE ON PRECEPTOR ROLES AND CLINICAL FACULTY ROLES IN THE NURSING WORKFORCE. CUR RENT PRECEPTORS IN THE COMMUNITY AND CLINICAL FACULTY ARE OFTEN ENGAGED IN THE ROLE OF PRECEPTOR WITHOUT FORMAL TRAINING AND LACK SUPPORT. THE PROJECT SEEKS TO REACH POPULATIONS THROUGH THE DEVELOPMENT OF EASILY ACCESSIBLE SELF-PACED INTERNET-BASED TRAINING MODULES, AN ANNUAL CLINICAL FACULTY AND PRECEPTOR CONFERENCE, AND INCORPORATION OF EMOTIONAL INTELLIGENCE AND RESILIENCY COACHING. THESE MODULES WILL COVER TOPICS IMPORTANT FOR CLINICAL FACULTY AND PRECEPTORS INCLUDING METHODS OF GIVING CONSTRUCTIVE FEEDBACK, UNCONSCIOUS BIAS, CHALLENGING STUDENT SITUATIONS, BUILDING STUDENT INDEPENDENCE AND CONFIDENCE, EMOTIONAL INTELLIGENCE, RESILIENCY, AND DEVELOPING CLINICAL STUDENT LEARNING AND EVALUATION PLANS. THE CLINICAL FACULTY AND PRECEPTOR ANNUAL CONFERENCE WILL BE HOSTED AT THE SIUE CAMPUS USING A HYBRID FORMAT TO FACILITATE ATTENDANCE FOR ALL PARTICIPANTS WITHIN HHS REGION 5. EACH YEAR THE PROGRAM’S GOAL IS TO TRAIN 60 STUDENTS IN GRADUATE NURSE EDUCATOR, NURSE ANESTHESIA, OR NURSE PRACTITIONER PROGRAMS, 100 HEALTHCARE PRECEPTORS AND 40 CLINICAL FACULTY FOR EACH YEAR OF THE PROJECT PROGRAM FUNDING. WHILE IDENTIFYING THE NEURAL UNDERPINNINGS OF ASD HAS BEEN A MAJOR FOCUS IN THE FIELD OF GENETICS AND NEUROSCIENCE, IT IS NOT CLEAR WHICH BRAIN REGION (S) AND CELL TYPES MAY BE FUNCTIONING ABNORMALLY TO GIVE RISE TO ASD. THE CEREBELLUM, A BRAIN REGION TYPICALLY CONSIDERED TO BE INVOLVED IN MOTOR COORDINATION AND CONTROL, HAS RECEIVED CONSIDERABLE ATTENTION FOR ITS POTENTIAL ROLE IN ASD, WITH A GROWING BODY OF CLINICAL EVIDENCE CORRELATING ASD DIAGNOSES WITH ABNORMALITIES IN CEREBELLAR ANATOMY, FUNCTION, AND MOTOR OR VESTIBULAR BEHAVIORS/FUNCTIONS. IN PARALLEL, CEREBELLAR INVOLVEMENT IN CIRCUITS OUTSIDE OF THE MOTOR SYSTEM PROVIDES A SUBSTRATE FOR THE CEREBELLUM TO INFLUENCE NON-MOTOR BEHAVIORS AND PROCESSES, SETTING THE STAGE FOR THE IMPORTANCE OF APPROPRIATE CEREBELLAR FUNCTION IN A HOST OF NEURAL PROCESSES. WITHIN THE CEREBELLUM, THE INITIAL INTEGRATION OF ALL INCOMING SENSORIMOTOR INFORMATION ENTERING THE CEREBELLAR CORTEX IS CARRIED OUT BY THE MORPHOLOGICALLY SIMPLE AND EXTREMELY DENSE POPULATION OF GRANULE CELLS. HOWEVER, LITTLE IS KNOWN ABOUT THE ROLE AND IMPORTANCE OF MANY ASD-LINKED GENES IN THE CEREBELLUM, ESPECIALLY IN GRANULE CELLS. IT IS ALSO NOT CLEAR WHAT ASPECTS OF CEREBELLAR GRANULE CELL FUNCTION AND CONNECTIVITY MAY BE IMPORTANT FOR SHAPING CERTAIN NON-MOTOR (AND MOTOR) BEHAVIORS. TO ADDRESS THIS KNOWLEDGE GAP, IN AIMS 1 AND 2 WE PROPOSE TO IDENTIFY THE DEGREE TO WHICH AN ASD-LINKED GENE DETERMINES PROPERTIES OF CEREBELLAR GRANULE CELL SYNAPTIC INTERACTIONS AND CORTICAL CIRCUITRY DYNAMICS OVER TIME. IN AIM 3, WE WILL IDENTIFY HOW A PARTICULAR ASD-LINKED GENE IN CEREBELLAR GRANULE CELLS INFLUENCE BEHAVIORAL PHENOTYPE ACROSS MOTOR AND NON-MOTOR DOMAINS. CONVERGING EARTH SCIENCE AND SUSTAINABILITY EDUCATION AND EXPERIENCE TO PREPARE NEXT-GENERATION GEOSCIENTISTS -THIS PROJECT WILL CONTRIBUTE TO THE NATIONAL NEED FOR WELL-EDUCATED SCIENTISTS, MATHEMATICIANS, ENGINEERS, AND TECHNICIANS BY SUPPORTING THE RETENTION AND GRADUATION OF HIGH-ACHIEVING, LOW-INCOME STUDENTS WITH DEMONSTRATED FINANCIAL NEED AT SOUTHERN ILLINOIS UNIVERSITY CARBONDALE (SIUC). SIUC IS A RURAL UNIVERSITY AND THE MAJOR RESEARCH INSTITUTION IN SOUTHERN ILLINOIS, ONE OF AMERICA?S MOST ECONOMICALLY DISTRESSED REGIONS. OVER ITS FIVE-YEAR DURATION, THIS PROJECT WILL FUND SCHOLARSHIPS TO 40 UNIQUE FULL-TIME TRANSFER STUDENTS WHO ARE PURSUING BACHELOR?S DEGREES IN GEOGRAPHY & ENVIRONMENTAL RESOURCES AND GEOLOGY. TRANSFER STUDENTS WILL RECEIVE TWO-YEAR SCHOLARSHIPS. THE PROPOSED PROJECT IS AN INTERDISCIPLINARY STEM PROGRAM, EARTH-SUSTAINABILITY SCHOLARSHIP (ESS), FOR ACADEMICALLY TALENTED STUDENTS WHO DEMONSTRATE FINANCIAL NEED TO PURSUE UNDERGRADUATE ENVIRONMENTAL AND EARTH SCIENCE DEGREES. A SUSTAINABILITY-MINDED STEM WORKFORCE IS VITAL FOR THE NATION TO STRATEGICALLY DEVELOP NATURAL RESOURCES, EXPERIENCE ECONOMIC GROWTH, AND MAKE INFORMED DECISIONS IN A RAPIDLY CHANGING WORLD. THE PROJECT WILL DEVELOP AN EDUCATIONAL PIPELINE FROM STUDENT ADMISSION AND TRAINING TO GRADUATION AND PROFESSIONAL DEVELOPMENT AND ADVANCE THE UNDERSTANDING OF THE DOMINANT FACTORS THAT INCREASE THE ACADEMIC AND PROFESSIONAL SUCCESS OF LOW-INCOME STUDENTS. A MENTORING SYSTEM AND A NETWORKED SUPPORTING COMMUNITY THAT INTEGRATE CAMPUS RESOURCES AND SUSTAINABILITY ACTIVITIES WILL BE ESTABLISHED FOR IMPROVED CURRICULAR AND CO-CURRICULAR EXPERIENCES. THE PROJECT IS EXPECTED TO BROADEN THE PARTICIPATION OF LOW-INCOME STUDENTS IN ENVIRONMENTAL AND EARTH SCIENCE BY REDUCING FINANCIAL PRESSURE, INCREASING LEARNING OPPORTUNITIES AND OUTCOMES, AND BRIDGING OVERALL CAREER SUCCESS. IT WILL ALSO ADVANCE THE MISSION OF U.S. INSTITUTIONS OF HIGHER EDUCATION THAT TRAIN A DIVERSE AND GLOBALLY COMPETITIVE STEM WORKFORCE IN SUSTAINABLE ENVIRONMENTAL AND EARTH SCIENCE. THE OVERALL GOAL OF THIS PROJECT IS TO INCREASE STEM DEGREE COMPLETION OF LOW-INCOME, HIGH-ACHIEVING UNDERGRADUATES WITH DEMONSTRATED FINANCIAL NEED. SPECIFIC OBJECTIVES INCLUDE: (1) INCREASING EDUCATIONAL OPPORTUNITIES FOR ACADEMICALLY TALENTED STUDENTS WITH FINANCIAL NEED, (2) IMPROVING EDUCATIONAL SUPPORT PROGRAMS AT SIUC, (3) IMPROVING THE RETENTION OF STUDENTS IN THE SIUC SCHOOL OF EARTH SYSTEMS AND SUSTAINABILITY, AND (4) ENHANCING THE FUTURE STEM WORKFORCE WITH SKILLS IN EARTH SCIENCE AND SUSTAINABILITY. TO ADDRESS THE HISTORIC DISCONNECT BETWEEN SUSTAINABILITY AND EARTH SCIENCE TRAINING, IT IS VITAL TO INTEGRATE SUSTAINABILITY INTO EARTH SCIENCE EDUCATION IN INSTITUTES OF HIGHER EDUCATION. ADVANCING UNDERSTANDING OF HOW SUPPORTING SERVICES AND PROGRAMS DRIVE ACADEMIC SUCCESS IN GEOSCIENCE MAJORS, ESPECIALLY FOR TRANSFER STUDENTS, IS ALSO CRITICAL. THIS PROJECT WILL ESTABLISH AN EDUCATIONAL PROTOTYPE THAT SUCCESSFULLY SUPPORTS ACADEMICALLY TALENTED AND LOW-INCOME TRANSFER STUDENTS TO BECOME SUSTAINABILITY-FOCUSED EARTH SCIENCE DEGREE GRADUATES. FOLLOWING THE THEORY OF PLANNED BEHAVIORS, THE SUPPORTING SERVICE AND ACTIVITIES TO ENHANCE ATTITUDES, SUBJECTIVE NORMS, AND PERCEIVED BEHAVIORAL CONTROL WILL BE FRAMED AS THREE CRITICAL CONSTRUCTS THAT ARE EXPECTED TO DRIVE STUDENTS? POSITIVE INTENTIONS FOR GRADUATION. THE PROJECT HOPES TO CREATE AN INCLUSIVE AND DIVERSE LEARNING COMMUNITY AND INFRASTRUCTURE TO ADVANCE THE PROJECT GOALS AND MISSIONS. IT IS EXPECTED THAT 40 LOW-INCOME ACADEMICALLY TALENTED TRANSFER STUDENTS WILL BE AWARDED $10,000 SCHOLARSHIPS EACH YEAR AND THAT AN OVERALL 85% RETENTION RATE AND 80% GRADUATION RATE WILL BE ACHIEVED. THE PROJECT WILL UTILIZE A MANAGEMENT-ORIENTED FORMATIVE AND SUMMATIVE EVALUATION APPROACH. RESULTS WILL BE DISSEMINATED INTERNALLY TO THE OFFICE OF THE PROVOST AND FACULTY MEMBERS AT SIUC AND SHARED EXTERNALLY TO THE STEM COMMUNITY THROUGH STEM CENTRAL, SOCIAL MEDIA, PROFESSIONAL CONFERENCES, AND PUBLICATIONS IN STEM EDUCATION JOURNALS. THIS PROJECT IS FUNDED BY NSF?S SCHOLARSHIPS IN SCIENCE, TECHNOLOGY, ENGINEERING, AND MATHEMATICS PROGRAM, WHICH SEEKS TO INCREASE THE NUMBER OF LOW-INCOME ACADEMICALLY TALENTED STUDENTS WITH DEMONSTRATED FINANCIAL NEED WHO EARN DEGREES IN STEM FIELDS. IT ALSO AIMS TO IMPROVE THE EDUCATION OF FUTURE STEM WORKERS, AND TO GENERATE KNOWLEDGE ABOUT ACADEMIC SUCCESS, RETENTION, TRANSFER, GRADUATION, AND ACADEMIC/CAREER PATHWAYS OF LOW-INCOME STUDENTS. COLLABORATIVE RESEARCH: DEVELOPING STEM EDUCATION REFORM LEADERS THROUGH A MULTIDISCIPLINARY AND CROSS-INSTITUTIONAL COMMUNITY OF TRANSFORMATION -THIS PROJECT AIMS TO SERVE THE NATIONAL INTEREST BY SYSTEMATICALLY SCALING UP EVIDENCE-BASED TEACHING PRACTICES IN SCIENCE, TECHNOLOGY, ENGINEERING, AND MATHEMATICS (STEM) COURSES ACROSS A DIVERSE SET OF INSTITUTIONS AND BY INVESTIGATING FACTORS ASSOCIATED WITH EFFECTIVE EDUCATIONAL INTERVENTIONS. TO UNDERSTAND AND ASSESS THESE FACTORS, THE COLLABORATIVE RESEARCH PROJECT INTENDS TO RIGOROUSLY IMPLEMENT AND INVESTIGATE A RECOGNIZED EFFECTIVE TEACHING PRACTICE, FLIPPED INSTRUCTION, IN WHICH STUDENTS APPLY KNOWLEDGE FROM WELL-STRUCTURED MATERIALS TO ACTIVE LEARNING SITUATIONS DURING CLASS TIME. A TEAM OF FACULTY AND RESEARCHERS FROM SOUTHERN ILLINOIS UNIVERSITY - EDWARDSVILLE, NORTHEASTERN ILLINOIS UNIVERSITY, SOUTHERN ILLINOIS UNIVERSITY CARBONDALE, AND CHICAGO STATE UNIVERSITY AND THEIR COMMUNITY COLLEGE PARTNERS, ST. LOUIS COMMUNITY COLLEGE, CITY COLLEGES OF CHICAGO-MALCOLM X COLLEGE, OAKTON COLLEGE, AND SHAWNEE COMMUNITY COLLEGE INTEND TO CONDUCT A WIDE-RANGE OF PROFESSIONAL DEVELOPMENT ACTIVITIES. THEY ALSO AIM TEST A MODEL OF PROFESSIONAL DEVELOPMENT, AND DETERMINE THE EFFECTS OF INSTITUTIONAL, DISCIPLINARY, INSTRUCTOR, AND STUDENT CHARACTERISTICS ON THE EFFECTIVENESS OF FLIPPED INSTRUCTION IN STEM UNDERGRADUATE EDUCATION. FOUR PAIRINGS OF INSTITUTIONS - EACH PAIR INCLUDING A 4-YEAR INSTITUTION AND A 2-YEAR INSTITUTION - CONSTITUTE THE FOUNDATION FOR BOTH IMPLEMENTATION AND INVESTIGATION. FLIPPED INSTRUCTION HAS DEMONSTRATED ITS POTENTIAL TO SIGNIFICANTLY TRANSFORM TEACHING AND LEARNING IN UNDERGRADUATE STEM EDUCATION. DRAWING ON EXTENSIVE RESEARCH AND FINDINGS FROM A PRIOR NSF IUSE EXPLORATORY AWARD, THIS TRACK 2 INSTITUTIONAL AND COMMUNITY TRANSFORMATION LEVEL II COLLABORATIVE RESEARCH PROJECT INTENDS TO (1) MONITOR STUDENT- CENTERED ORIENTATION OF FACULTY, IMPLEMENTATION, AND VARIATIONS IN IMPLEMENTATION BASED ON INSTITUTIONAL TYPE, DEPARTMENT/DISCIPLINE, AND STUDENT DEMOGRAPHICS; (2) IDENTIFY ENABLERS AND OBSTACLES; AND (3) EXAMINE CHANGE LEADERSHIP AS FOUNDATIONAL STRATEGIES FOR DEVELOPING A COMMUNITY OF PRACTICE. ACTIVITIES HAVE BEEN DESIGNED TO ENHANCE INSTRUCTOR EXPERTISE AND SKILLS IN FLIPPED INSTRUCTION TO MEET THE NEEDS OF STUDENTS AND LEADERSHIP TO DRIVE TRANSFORMATIVE, LASTING CHANGE WITHIN THE RESPECTIVE INSTITUTIONS. GROUNDED IN A COMMUNITIES OF TRANSFORMATION PERSPECTIVE, THE MIXED METHODS RESEARCH DESIGN USES RIGOROUS DATA COLLECTION, ANALYSIS, AND INTERPRETATION METHODS. THE TEAM WILL DOCUMENT AND ASSESS INSTRUCTORS' CONFIDENCE, ATTITUDES, AND MOTIVATIONS TO USE FLIPPED INSTRUCTION, AS WELL AS STUDENTS' ACADEMIC PERFORMANCE AND PERSISTENCE IN STEM. THE INNOVATIVE COLLABORATION OF EIGHT INSTITUTIONS (FOUR PAIRS) INCLUDES INTENTIONALLY DESIGNED TRAINING FOR EFFECTIVE STEM TEACHING, WHILE THE RESEARCH SEEKS TO ADDRESS DIFFERENTIAL EFFECTS ACROSS INSTITUTIONS AND FIND SOLUTIONS THAT STRENGTHEN STEM EDUCATION. THE PROJECT WILL IDENTIFY ENABLERS AND CHALLENGES TO LASTING CHANGE IN UNDERGRADUATE STEM EDUCATION, BOTH AT THE FOUR-YEAR AND COMMUNITY COLLEGE LEVELS. THE NSF IUSE: EDU PROGRAM SUPPORTS RESEARCH AND DEVELOPMENT PROJECTS TO IMPROVE THE EFFECTIVENESS OF STEM EDUCATION FOR ALL STUDENTS. THROUGH THE INSTITUTIONAL AND COMMUNITY TRANSFORMATION TRACK, THE PROGRAM SUPPORTS EFFORTS TO TRANSFORM AND IMPROVE STEM EDUCATION ACROSS INSTITUTIONS OF HIGHER EDUCATION AND DISCIPLINARY COMMUNITIES. SINGLE-MOLECULE APPROACHES TO STUDY EPIBLAST STEM CELL FATE DECISION - PROJECT SUMMARY TISSUE AND ORGAN FAILURE, EITHER DUE TO INJURY OR AGING, ARE BECOMING A MAJOR HEALTH PROBLEM WORLDWIDE WITH AN ESTIMATED COST OF ONE-HALF OF THE TOTAL ANNUAL HEALTHCARE EXPENSES. TO ADDRESS THIS ISSUE, TISSUE ENGINEERING APPROACHES CAN BE LEVERAGED BY UTILIZING FUNCTIONAL BODY CELLS CREATED IN A LABORATORY SETTING. PLURIPOTENT EPIBLAST STEM CELLS (EPISCS) CAN SERVE AS AN EXCELLENT MODEL TO DETERMINE HOW TO DIRECT CELL FATE FOR CREATING FUNCTIONAL BODY CELLS. HOWEVER, EVEN WITH THE BEST CHEMICALLY-DEFINED DIFFERENTIATION PROTOCOL OF PLURIPOTENT STEM CELLS, THE CONTROL OF CELL-LINEAGE SPECIFICATION REMAINS POOR. BESIDES CHEMICAL SIGNALING, IT IS NOW WIDELY ACCEPTED THAT PHYSICAL SIGNALS FROM THE EXTRACELLULAR MATRIX (ECM) PLAY A CRUCIAL ROLE IN CELL FATE DETERMINATION. NEVERTHELESS, CONTROL OF CELL-LINEAGE SPECIFICATION BY SUCH MECHANICAL FORCES ALONE COULD NOT BE IMPROVED POSSIBLY DUE TO THE LACK OF PRECISE CONTROL OF FORCES AT THE SINGLE-MOLECULE LEVEL AND THE LACK OF SYNERGY BETWEEN CHEMICAL SIGNALING AND MECHANICAL PATHWAYS. TO ADDRESS THIS GAP, THE PROPOSED STUDY AIMS TO PROVIDE A MECHANISTIC FRAMEWORK OF SINGLE EPISC FATE DECISIONS (SELF-RENEWAL AND DIFFERENTIATION) BASED ON CHEMICAL AND SINGLE-MOLECULE FORCE BASED APPROACHES. THE CENTRAL HYPOTHESIS IS THAT THE SYNERGISTIC EFFECT OF CHEMICAL AND SINGLE-MOLECULE FORCE CUES VIA CELL-ECM AND CELL-CELL INTERACTIONS CAN CONTROL FATE DECISIONS FAR MORE EFFECTIVELY THAN PREVIOUSLY POSSIBLE. THE LONG-TERM GOAL IS TO DEVELOP NOVEL APPROACHES TO CONTROL THE DIRECTED DIFFERENTIATION OF PLURIPOTENT CELLS INTO ALL THREE GERM-LAYERS. TO THIS END, THE FOLLOWING THREE AIMS ARE PROPOSED. SPECIFICALLY, AIM 1 WILL FOCUS ON UNDERSTANDING THE MECHANISM OF SINGLE-MOLECULE FORCE MEDIATED DIFFERENTIATION OF EPISCS INTO THE MESODERM LINEAGE. THE FORCE TRANSMISSION INTO SINGLE CELLS VIA SINGLE AVSS3 INTEGRINS WILL BE CONTROLLED BY TENSION GAUGE TETHERS. THESE DNA-BASED RUPTURABLE TETHERS CAN PRECISELY LIMIT THE AMOUNT OF FORCE AT THE SINGLE-MOLECULE LEVEL. TOGETHER WITH CHEMICAL SIGNALING, SUCH PRECISE CONTROL AND SPECIFIC TARGETING OF MECHANICAL PATHWAYS MAY LEAD TO SUPERIOR CONTROL OF CELL DIFFERENTIATION INTO THE MESODERM. IN AIM 2, THE MECHANISM OF SELF-RENEWAL OF SINGLE EPISCS WILL BE IDENTIFIED BY DEFINING A MICROENVIRONMENT COMPOSED OF SELF-RENEWAL PROMOTING LIGANDS SUCH AS E-CADHERIN. IN AIM 3, DIFFERENTIATION OF SINGLE EPISCS WILL BE DEFINED VIA THE NOTCH PATHWAY BY ENGINEERED LOW- TOLERANCE TENSION GAUGE TETHER CALLED “NANO-YOYO” TO ACTIVATE FORCE-DEPENDENT NOTCH SIGNALING. EVIDENCE SHOWS THAT STUDENTS FROM RURAL AREAS OFTEN RETURN TO THEIR OWN COMMUNITIES UPON DEGREE COMPLETION, MAKING IT CRUCIAL TO RECRUIT AND RETAIN STUDENTS FROM THESE LOCATIONS. LIMITED FINANCIAL RESOURCES AND LACK OF KNOWLEDGE ABOUT PATHWAYS TO NURSING DEGREE COMPLETION CONTRIBUTE TO THESE SHORTAGES. ADDITIONALLY, THERE IS A NEED FOR INCREASED KNOWLEDGE IN LONG-TERM CARE, GERIATRIC SUPPORT, AND COMMUNITY NURSING TO BETTER SERVE RURAL AND ADJACENT POPULATIONS. THIS PROPOSED PROJECT HAS TWO SPECIFIC AIMS WITH SEVERAL OBJECTIVES TO ACHIEVE THESE AIMS SUCCESSFULLY. AIM 1: EXPAND NURSING WORKFORCE CAPACITY IN RURAL AND LONG-TERM CARE SETTINGS. OBJECTIVES TO ADDRESS AIM 1 INCLUDE: 1) COLLABORATE WITH COMMUNITY COLLEGES, UNIVERSITIES, AND CLINICAL PARTNERS IN RURAL AREAS FOR RECRUITMENT OUTREACH PROGRAMS IN MIDDLE AND HIGH SCHOOLS TO PROMOTE MULTIPLE PATHWAYS TO NURSING DEGREE COMPLETION. 2) IMPLEMENT INTENTIONAL RECRUITMENT EFFORTS FOCUSING ON DIVERSIFYING THE NURSING WORKFORCE. 3) PROVIDE FUNDING OPPORTUNITIES FOR STUDENTS IN THE ACCELERATED POST-BACCALAUREATE NURSING PROGRAM. AIM 2: ENHANCE EDUCATIONAL EXPERIENCES THROUGH TRAINING AND EXPERIENTIAL LEARNING ACTIVITIES FOCUSED ON RURAL AND LONG-TERM CARE READINESS. OBJECTIVES TO ADDRESS AIM 2 INCLUDE: 1) COLLABORATE WITH COMMUNITY COLLEGES, UNIVERSITIES, AND CLINICAL PARTNERS IN RURAL AND LONG-TERM CARE SETTINGS TO PROVIDE CLINICAL ROTATIONS IN LONG-TERM CARE AND CRITICAL ACCESS HEALTHCARE DELIVERY CENTERS. 2) DEVELOP AND IMPLEMENT TRAINING PROGRAMS IN COLLABORATION WITH MULTIPLE CLINICAL PARTNERS AND COLLEGES THAT ENHANCE CRITICAL THINKING, UNDERSTANDING OF SOCIAL DETERMINANTS OF HEALTH, LEADERSHIP, AND COMMUNICATION SKILLS. 3) INCORPORATE VIRTUAL REALITY AND HANDS-ON SIMULATION EXPERIENCES INTO THE CURRICULUM WITH AN EMPHASIS ON COMMUNITY AND GERIATRIC CARE. BY ADDRESSING THESE CRITICAL NEEDS, THE PROJECT AIMS TO BUILD A ROBUST AND DIVERSE NURSING WORKFORCE EQUIPPED TO DELIVER HIGH-QUALITY HEALTHCARE IN RURAL AND UNDER-SERVED REGIONS. CELL-TYPES SPECIFIC NEUROADAPTATIONS IN THE NUCLEUS ACCUMBENS SHELL ASSOCIATED WITH INDIVIDUAL DIFFERENCES IN COCAINE-SEEKING - PROJECT SUMMARY/ABSTRACT ALTHOUGH COCAINE IS POWERFULLY REWARDING, NOT ALL PEOPLE WHO ARE EXPOSED TO THIS DRUG ARE EQUALLY PRONE TO ABUSING IT. THE REASONS FOR THESE DIFFERENCES REMAIN UNKNOWN BUT MAY INVOLVE PREDISPOSING FACTORS WHOSE ELUCIDATION COULD LEAD TO THERAPIES THAT, IN THE FUTURE, MAY PREVENT COCAINE ADDICTION. ONE SUCH PREDISPOSING FACTOR IS BEHAVIORAL SENSITIVITY TO THE AVERSIVE ATTRIBUTES OF COCAINE, WHICH OVERLAP WITH THE REWARDING EFFECTS TO PROTECT AGAINST COCAINE-SEEKING. PRELIMINARY DATA IN THIS PROPOSAL SUGGEST THAT THE AVERSIVE EFFECT OF COCAINE MAY BE MUCH STRONGER IN SOME INDIVIDUALS THAN OTHERS AND POTENTIALLY A DETERMINANT FOR COCAINE ADDICTION VULNERABILITY. THE UNDERLYING CELLULAR MECHANISM OF THIS VARIABILITY REMAINS UNKNOWN. HOWEVER, OUR PRELIMINARY DATA REVEALED THAT HIGHER RHOA ACTIVITY AND D2-MSN EXCITABILITY IN THE MNACSHELL CORRELATE WITH RATS DISPLAYING HIGHER AVERSIVE EFFECTS OF COCAINE (AND CONSEQUENTLY LOWER DRIVE TO SEEK COCAINE). NEVERTHELESS, IT IS UNKNOWN IF THESE CELLULAR AND MOLECULAR ADAPTATIONS IN THE MNACSHELL ARE INVOLVED IN THE AVERSIVE ATTRIBUTES OF COCAINE. ACCORDINGLY, TO ADDRESS THIS GAP IN KNOWLEDGE, I’M PROPOSING TWO AIMS. IN AIM 1, I WILL DETERMINE THE ROLES OF D2MSN ON COCAINE AVOIDANCE BEHAVIOR, AND FOR THIS, I WILL BE TRAINED IN FIBER PHOTOMETRIC AND OPTOGENETIC METHODS. I HYPOTHESIZE THAT D2- BUT NOT D1-MSNS ARE ACTIVATED DURING COCAINE’S AVERSIVE PHASE, AND THAT D2 ACTIVITY IN TURN DRIVES CONDITIONED NEGATIVE ATTRIBUTES OF COCAINE. IN AIM 2, I WILL EXAMINE THE ROLES OF RHOA AND RELATED GENES ON NEURAL EXCITABILITY AND COCAINE AVOIDANCE IN A CELL-SPECIFIC MANNER. FIRST, I WILL USE MRNA TRAP METHODS TO ANALYZE CELL-SPECIFIC GENE TRANSLATION ASSOCIATED WITH RHOA SIGNALING OR EXCITABILITY LINKED TO COCAINE AVOIDANCE PHENOTYPES. IN ADDITION, TO TEST THE CAUSAL ROLE OF RHOA IN EXCITABILITY AND COCAINE AVOIDANCE BEHAVIOR, I WILL DECREASE OR INCREASE RHOA ACTIVITY IN A CELL-SPECIFIC MANNER USING AN ADENO-ASSOCIATED GENE MANIPULATION STRATEGY. I HYPOTHESIZE THAT INCREASES IN RHOA ACTIVITY IN D2-MSN BUT NOT D1-MSN ENHANCES EXCITABILITY IN THE MNACSHELL CONFER PROTECTION AGAINST THE ACQUISITION OF COCAINE-SEEKING. AN INTENSE AND COMPREHENSIVE TRAINING, MENTORING, AND RESEARCH PLAN HAS BEEN DEVELOPED TO ACHIEVE THESE GOALS. THIS PLAN WILL BE GUIDED BY A POWERFUL TEAM OF MENTORS AND COLLABORATORS THAT WILL HELP BLEND MY CURRENT SKILL SET WITH NEW CONCEPTUAL AND TECHNICAL FRAMEWORKS AS I CONTINUE TO ESTABLISH MY OWN UNIQUE AND INNOVATIVE RESEARCH PROGRAM IN ADDICTION NEUROSCIENCE.

AIM 1: RECRUITMENT OF PROSPECTIVE STUDENTS AND NURSE EDUCATORS WITH AN EMPHASIS ON DIVERSIFYING THE NURSING WORKFORCE. AIM 2: RETENTION OF PRE-NURSING AND NURSING STUDENTS AT A STATE PUBLIC UNIVERSITY. AIM 3: READINESS TO PRACTICE THROUGH A PROGRAM FOCUSING ON ENHANCING THE UNDERGRADUATE NURSING PROGRAM EXPERIENCE TO ENSURE COMPETENCY AND CONFIDENCE IN PRACTICE ENTRY IN ACUTE CARE SETTINGS UPON GRADUATION. EVIDENCE HAS SHOWN THAT CLIENT TRUST AND PATIENT OUTCOMES ARE IMPROVED WHEN HEALTH CARE PROFESSIONALS ARE REPRESENTATIVE OF THE NATURAL VARIATIONS WITHIN THE POPULATION. THERE IS ALSO AN ONGOING, SIGNIFICANT NURSING SHORTAGE MAKING IT IMPERATIVE THAT ACADEMIC AND CLINICAL COLLABORATIONS ARE IN PLACE TO ENSURE READINESS TO PRACTICE FOR NEW GRADUATE NURSES. EVIDENCE SHOWS THAT CLINICAL, CLASSROOM AND SIMULATION EXPERIENCES THAT EMPHASIZE CONCEPTS FROM SOCIAL DETERMINANTS OF HEALTH PREPARE UNDERGRADUATE NURSES FOR APPROPRIATE AND INCLUSIVE CARE OF DIVERSE POPULATIONS. IN ADDITION, LITERATURE IS ABUNDANT WITH STRATEGIES TO RECRUIT AND RETAIN NURSES OF DIVERSE BACKGROUNDS WHICH INCLUDES MENTORING, ACADEMIC, SOCIAL AND PERSONAL ADVISING, AND SUPPORTIVE ACADEMIC ENVIRONMENTS. OBJECTIVES OF THIS PROJECT INCLUDE: 1) RECRUITMENT OF DIVERSE NURSING STUDENTS THROUGH DEVELOPMENT OF A SCHOOL PIPELINE IN COLLABORATION WITH SCHOOL DISTRICTS AND LOCAL HOSPITALS TO FACILITATE AWARENESS OF THE NURSING PROFESSION BY OFFERING SHADOWING EXPERIENCES AND MOBILE SIMULATION EXPERIENCES. ADDITIONAL EFFORTS TO INCREASE DIVERSE NURSING FACULTY INCLUDE NATIONAL RECRUITMENT EFFORTS AND DEVELOPMENT OF NURSE EDUCATOR FELLOWSHIPS TO ASSIST IN DEGREE COMPLETION, RESEARCH EFFORTS OR FACULTY DEVELOPMENT GOALS; 2) RETENTION OF PRE-NURSING STUDENTS THROUGH COLLABORATION WITH OUR SCIENCE DEPARTMENT TO INTEGRATE AN EVIDENCE-BASED PEER LED TEAM LEARNING MODEL THAT IS WELL-KNOWN TO IMPACT STUDENT SUCCESS; 3) RETENTION OF STUDENTS BY INCREASING RESOURCES FOR OUR STUDENT SUCCESS TEAM WITHIN THE NURSING PROGRAM AS WELL AS OUR ALREADY ESTABLISHED AND SUCCESSFUL STUDENT NURSE ACHIEVEMENT PROGRAM (SNAP); 4) ENHANCED READINESS TO PRACTICE THROUGH COLLABORATIONS WITH HOSPITALS LOCATED IN UNDERSERVED AREAS TO FOSTER INNOVATIVE CLINICAL EXPERIENCES THAT INVOLVE A NURSE PRECEPTOR MODEL.
