NIHL HAS A HIGH PREVALENCE AMONG MEMBERS OF THE ARMED FORCES AND FIREFIGHTERS WHO ARE OCCUPATIONALLY AT-RISK. GLOBALLY, OCCUPATIONAL NOISE EXPOSURE IS RESPONSIBLE FOR 16% OF DISABLING HEARING LOSS (OVER 600 MILLION PEOPLE) WITH SIGNIFICANT LIFETIME COSTS PER PERSON IN THE US ALONE. PRELIMINARY AND PRIOR PUBLISHED RESULTS SHOW THAT CONTROLLED AND LOCALIZED THERAPEUTIC HYPOTHERMIA PROVIDED TO THE INNER EAR POST-NOISE OR SURGICAL TRAUMA CONSERVES SIGNIFICANT RESIDUAL HEARING AND PRESERVES SENSITIVE NEURAL STRUCTURES. OVER THE PAST 3 YEARS, THE UNIVERSITY OF MIAMI TEAM HAS ALSO SHOWN NOISE- INDUCED CHANGES IN HEARING AND BALANCE FUNCTIONS IN AT-RISK GROUPS OF FIREFIGHTERS. WITH THE PRIOR PHASE I SBIR AWARD, RESTOREAR DEVICES LLC DESIGNED, BUILT AND TESTED SYSTEMS AND PROTOCOLS TO DELIVER MILD THERAPEUTIC HYPOTHERMIA NON-INVASIVELY TO THE INNER EAR. WE MET ALL OF THE MILESTONES OUTLINED, INCLUDING TESTING THE EFFICACY OF OUR DEVICES FOR HUMAN APPLICATION USING COMPUTATIONAL MODELS OF HEAT TRANSFER AND STUDIES FROM CADAVERIC TEMPORAL BONES. WE ARE NOW READY TO TRANSLATE ONE SUCH DEVICE, REBOUNDTM, TO PRACTICE. OUR VISION WITH THE INFORMED BIOBANKING PROGRAM WAS TO DEVELOP A SCALABLE BIOBANKING WORKFLOW THAT ENSURES ALL U.S. ENDANGERED SPECIES CAN BE BIOBANKED AS PART OF LONG-TERM RECOVERY STRATEGIES. FOR AMPHIBIANS AND REPTILES, THE GOAL WILL BE TO BIOBANK LIVING TISSUES (AKA EXPRESS TISSUE BANKING) FOR 1 FEMALE AND 1 MALE, AND ALSO STORE THOSE TISSUES LONG-TERM AT THE USDA GERMPLASM BIOREPOSITORY. WE RECOGNIZE IT MAY NOT BE POSSIBLE TO COLLECT TISSUES FROM ALL THE TAXA LISTED IN TABLE 1 OVER THE COURSE OF A SINGLE YEAR DUE TO THE RARITY AND ELUSIVENESS OF SOME TAXA, AS WELL AS UNFORESEEN CIRCUMSTANCES THAT MAY IMPEDE COLLECTION (E.G. LIVING SHORELINES REFERS TO A VARIETY OF NATURE-BASED SHORELINE PROTECTION OPTIONS THAT ALLOW FOR NATURAL COASTAL PROCESSES TO REMAIN THROUGH THE STRATEGIC PLACEMENT OF PLANTS, STONE, SAND FILL, AND OTHER STRUCTURAL AND ORGANIC MATERIALS. LIVING SHORELINES BENEFIT COASTAL HABITATS, COMMUNITIES, AND ECOSYSTEMS ACROSS THE COUNTRY, AND THE COUNTLESS HUMANS AND SPECIES THAT LIVE WITHIN THEM. BENEFITS PROVIDED INCLUDE: 1. LIVING SHORELINES IMPROVE WATER QUALITY BY TRAPPING SEDIMENTS AND FILTERING POLLUTANTS. 2. LIVING SHORELINES PROVIDE SHALLOW WATER HABITAT AND A DIVERSITY OF PLANT SPECIES FOR AQUATIC AND TERRESTRIAL ANIMALS THEY ALSO PROVIDE SHADE TO KEEP WATER TEMPERATURES COOL, HELPING TO INCREASE OXYGEN LEVELS FOR FISH AND OTHER AQUATIC SPECIES. 3. THE PURPOSE OF THE AWARD IS TO IMPROVE ENVIRONMENTAL CONDITIONS IN LONG ISLAND SOUND AND CONCURRENTLY BUILD CAPACITY FOR COMMUNITIES AND INSTITUTIONS IN THE REGION. THESE EFFORTS WILL LEAD TO RESILIENT, SUSTAINABLE, THRIVING WATERSHEDS, AND STRENGTHEN ECONOMIC AND CULTURAL VIABILITY OF THE SOUND.ACTIVITIES:SPECIFICALLY, UNDER THIS COOPERATIVE AGREEMENT THE RECIPIENT WILL CARRY OUT TWO PRIMARY TASKS: 1. ADMINISTER A COMPETITIVE SUBAWARD PROGRAM FOCUSED ON ACTIVITIES THAT ADDRESS CHALLENGES AND OPPORTUNITIES FACING COMMUNITIES EXPERIENCING OR AFFECTED BY ENVIRONMENTAL AND HUMAN HEALTH RISKS OR HARMS. 2. PROVIDE TECHNICAL ASSISTANCE FOR COMMUNITIES AND SUPPORT PROPOSALS WHICH ULTIMATELY IMPROVE ACCESS TO THE LONG ISLAND SOUND, WHILE REDUCING OVERALL ENVIRONMENTAL HEALTH RISKS. SUBRECIPIENT:RAE WILL EXECUTE A SUBAWARDS PROGRAM BY MANAGING A COMPETITIVE PROCESS TO SOLICIT, EVALUATE, AND SELECT SUBAWARD PROJECTS THAT ADDRESS LISS GOALS AND PROGRAM OBJECTIVES TO SUPPORT CONNECTICUT AND NEW YORK COMMUNITIES, IMPLEMENT THE LISS CCMP, INCLUDING VARIETY OF PROJECT TYPES, DISTRIBUTION ACROSS THE LISS REGION, AND REPRESENTATION OF COMMUNITY NEEDS. OUTCOMES:UNDER THIS COOPERATIVE AGREEMENT THE RECIPIENT WILL PROVIDE A VARIETY OF DELIVERABLES RELATED TO THE TWO PRIMARY TASKS DESCRIBED ABOVE. WITHIN THE TECHNICAL ASSISTANCE PROGRAM, RESTORE AMERICA'S ESTUARIES WILL: CONVENE COMMUNITY ENGAGEMENT SESSIONS THROUGHOUT THE DURATION OF THE GRANT PROGRAM TO ADDRESS ISSUES COMMUNITIES ARE FACING THAT ARE RELEVANT FOR LONG ISLAND SOUND STUDY'S COMPREHENSIVE CONSERVATION AND MANAGEMENT PLAN PRIORITIES; EVOLVE TRAININGS BASED ON CONSTANT EVALUATIONS, AND ORGANIZATIONAL/REGIONAL NEEDS AS CAPACITY INCREASES; INCLUDE ADDITIONAL ANNUAL LEARNING EXCHANGES; CREATE 'MINI LEARNING EXCHANGES' IN CONNECTICUT AND NEW YORK THAT WILL COMPLEMENT THE MAIN ANNUAL LEARNING EXCHANGE IN A CENTRAL LOCATION. THE EXPECTED OUTCOMES OF THESE ACTIVITIES ARE IMPROVED ENVIRONMENTAL CONDITIONS IN THE LONG ISLAND SOUND WATERSHED AND STRENGTHENED ECONOMIC AND CULTURAL VIABILITY OF THE SOUND. LONG ISLAND SOUND COMMUNITY IMPACT FUND IS INTENDED TO INCREASE THE NUMBER OF COMMUNITY-LED PROJECTS THAT SUPPORT IMPLEMENTATION OF THE LONG ISLAND SOUND STUDY GOALS AND PRIORITIES LAID OUT IN ITS COMPREHENSIVE CONSERVATION AND MANAGEMENT PLAN; IT ALSO WILL BUILD CAPACITY IN COMMUNITIES EXPERIENCING OR AFFECTED BY ENVIRONMENTAL AND HUMAN HEALTH RISKS OR HARMS. DESCRIPTION:THE PURPOSE OF THIS AGREEMENT IS TO PROVIDE ASSISTANCE TO RESTORE AMERICA'S ESTUARIES TO IMPLEMENT ITS PROJECT TO SUPPORT THE LONG ISLAND SOUND COMPREHENSIVE CONSERVATION AND MANAGEMENT PLAN TO PROTECT AND RESTORE LONG ISLAND SOUND BY DEVELOPING AND ADMINISTERING THE LONG ISLAND SOUND COMMUNITY IMPACT FUND. THE FUND IS BEING CREATED TO INVOLVE COMMUNITIES EXPERIENCING OR AFFECTED BY ADVERSE AND DISPROPORTIONATE ENVIRONMENTAL AND HUMAN HEALTH RISKS OR HARMS, INCLUDING AFFECTED UNDERSERVED COMMUNITIES WITHIN THE SCOPE OF THE LONG ISLAND SOUND PLAN. ACTIVITIES:SPECIFICALLY, UNDER THIS COOPERATIVE AGREEMENT THE RECIPIENT WILL CARRY OUT TWO PRIMARY TASKS: 1) DEVELOP AND ADMINISTER A COMPETITIVE SUBAWARD PROGRAM FOCUSED ON ACTIVITIES THAT ADDRESS CHALLENGES AND OPPORTUNITIES FACING COMMUNITIES EXPERIENCING OR AFFECTED BY ADVERSE AND DISPROPORTIONATE ENVIRONMENTAL AND HUMAN HEALTH RISKS OR HARMS, INCLUDING AFFECTED UNDERSERVED COMMUNITIES WITHIN THE SCOPE OF THE LONG ISLAND SOUND COMPREHENSIVE CONSERVATION AND MANAGEMENT PLAN; 2) DEVELOP AND IMPLEMENT AN OUTREACH AND SUPPORT PROGRAM FOCUSED ON INVOLVING COMMUNITIES EXPERIENCING OR AFFECTED BY ADVERSE AND DISPROPORTIONATE ENVIRONMENTAL AND HUMAN HEALTH RISKS OR HARMS, INCLUDING AFFECTED UNDERSERVED COMMUNITIES AND PROVIDING THEM TECHNICAL SUPPORT AS NECESSARY. SUBRECIPIENT:RESTORE AMERICA'S ESTUARIES WILL EXECUTE A SUBAWARDS PROGRAM BY MANAGING A COMPETITIVE PROCESS TO SOLICIT, EVALUATE, AND SELECT SUBAWARD PROJECTS THAT ADDRESS LISS GOALS AND PROGRAM OBJECTIVES TO SUPPORT UNDERSERVED COMMUNITIES, INTEGRATE ENVIRONMENTAL JUSTICE INTO PROJECT IMPLEMENTATION AND IMPLEMENT THE LONG ISLAND SOUND PLAN, INCLUDING DIVERSITY OF PROJECT TYPES, DISTRIBUTION ACROSS THE REGION, AND REPRESENTATION OF THE NEEDS OF UNDERSERVED COMMUNITIES. RESTORE AMERICA'S ESTUARIES WILL PERFORM SUBAWARDEE MONITORING, INCLUDING PROGRAMMATIC AND FINANCIAL OVERSIGHT, THROUGH SEMI-ANNUAL PROGRESS REPORTS, INVOICE REVIEWS, AND SITE VISITS TO ENSURE PERFORMANCE ACCOUNTABILITY AND WILL ENSURE SUBAWARDS WILL BE COMPLETED PRIOR TO THE END OF THE COOPERATIVE AGREEMENT. THERE IS $3,000,000 ALLOCATED TO THIS SUBAWARD PROGRAM. RESTORE AMERICA'S ESTUARIES ALSO IS PROVIDING A $54,603 SUBAWARD TO SAVE THE SOUND. THE PURPOSE OF THIS SUBAWARD IS FOR SAVE THE SOUND TO SUPPORT OUTREACH TO LONG ISLAND SOUND COMMUNITIES WITH ENVIRONMENTAL JUSTICE CONCERNS. OUTCOMES:UNDER THIS COOPERATIVE AGREEMENT THE RECIPIENT WILL PROVIDE A VARIETY OF DELIVERABLES RELATED TO THE TWO PRIMARY TASKS DESCRIBED ABOVE. THE SUBAWARD PROGRAM DELIVERABLES INCLUDE: COLLABORATIVE SOLICITATION DEVELOPMENT, TARGETED OUTREACH TO RELEVANT GROUPS THAT CAN BENEFIT MOST FROM THE PROGRAM, ESTABLISHMENT OF TRANSPARENT, EQUITABLE AND INCLUSIVE EVALUATION CRITERIA AND REVIEW PANELS AND SUBAWARD OVERSIGHT/MANAGEMENT. WITHIN THE OUTREACH AND SUPPORT PROGRAM, RESTORE AMERICA'S ESTUARIES WILL DELIVER: IDENTIFICATION OF HIGH POTENTIAL ORGANIZATIONS RELEVANT TO THE FUND, ORGANIZATION OF GRANT MAKING WORKSHOP FOR POTENTIAL SUB-AWARDEES, MENTORSHIP AND CONNECTION THROUGHOUT THE APPLICATION PROCESS, DETAILED FEEDBACK TO ALL APPLICANTS. THE FIRST INSTALLMENT OF THIS INCREMENTAL AWARD ($2,364,527) WILL COVER DELIVERABLES FOR YEARS 1 AND 2 OUTLINED IN THE PROJECT NARRATIVE AND BUDGET NARRATIVE PROVIDED BY RESTORE AMERICA'S ESTUARIES (ATTACHMENTS INCLUDED IN SECTION M). FUNDING FOR DELIVERABLES COVERED IN YEARS 3-5 WILL BE MADE THROUGH ADDITIONAL INCREMENTAL AWARDS TO THE RECIPIENT. THE EXPECTED OUTCOMES OF THESE ACTIVITIES ARE IMPROVED ENVIRONMENTAL CONDITIONS IN THE LONG ISLAND SOUND WATERSHED AND STRENGTHENED ECONOMIC AND CULTURAL VIABILITY OF LONG ISLAND SOUND WATERSHED COMMUNITIES. RESTORE AMERICA'S ESTUARIES WILL BE AWARDED $4,993,897 TO RESTORE OYSTER REEF HABITAT AT SITES ACROSS THE GULF OF MEXICO REGION. OYSTER SHELL RECYCLING PROGRAMS WILL BE EXPANDED AND ENHANCED AROUND THE GULF. SHELLS WILL BE COLLECTED FROM RESTAURANTS, RECYCLED, AND PUT BACK IN THE ENVIRONMENT AT OYSTER REEF RESTORATION SITES. SHELL RECYCLING PROGRAMS AND OYSTER REEF RESTORATION SITES WILL BE DESIGNED TO SERVE LOCAL ECOSYSTEM AND COMMUNITY RESILIENCE NEEDS, WITH AN EMPHASIS ON TRIBES AND UNDERSERVED COMMUNITIES. THE RESTORED OYSTER REEFS WILL PROVIDE HABITAT FOR A DIVERSE GROUP OF SPECIES, INCLUDING RECREATIONALLY AND COMMERCIALLY IMPORTANT FISH AND THEIR PREY. DEVICE TO DELIVER INTRA-OPERATIVE THERAPEUTIC HYPOTHERMIA FOR HEARING PRESERVATION IN COCHLEAR IMPLANTATION - PROJECT SUMMARY THE GOAL OF OUR PROJECT IS TO FINALIZE, VALIDATE, AND PREPARE FOR REGULATORY APPROVALS A DEVICE AND PROBE FOR DELIVERING LOCALIZED, CONTROLLED THERAPEUTIC HYPOTHERMIA TO THE INNER EAR. THE DEVICE SAFETY AND EFFICACY WILL BE TESTED FOR RESIDUAL HEARING PRESERVATION IN COCHLEAR IMPLANT SURGERIES BY COMPARING PRE- AND POST-OPERATIVE HEARING OUTCOMES AND INTRA-OPERATIVE ELECTROPHYSIOLOGICAL MEASUREMENTS. EXTENSIVE PRECLINICAL RESEARCH WAS ENABLED BY A NIDCD R21 AND INDUSTRY SUPPORT THAT ALLOWED DEVELOPMENT OF A PROTOCOL FOR HYPOTHERMIA DELIVERY USING A DEVICE AND PROBE SUITABLE FOR A RODENT MODEL. WE PROPOSE HERE TO RE-ENGINEER THE DEVICE AND PROBE FOR CLINICAL IMPLANTATION IN THE OPERATING ROOM, AND PREPARE IT FOR REGULATORY APPROVAL. WE WILL ALSO CARRY OUT FIRST-IN- PATIENT STUDIES FOR THE THERAPY BY IMPLEMENTING LOCALIZED, THERAPEUTIC HYPOTHERMIA FOR PROTECTION OF HAIR CELLS AND NEURAL SUBSTRATE DURING COCHLEAR IMPLANTATION. THIS APPROACH BRINGS A NON-PHARMACEUTICAL THERAPY FOR RESIDUAL HEARING PRESERVATION TO THE MARKET. FURTHERMORE, HYPOTHERMIA LOCALIZED TO THE INNER EAR AVOIDS CHALLENGES WITH SYSTEMIC COOLING OR WITH DELIVERY OF DRUGS TO THE INNER EAR. THE DEVICE AND PROBE DO NOT REQUIRE ALTERATION OF THE SURGICAL APPROACH OR THE IMPLANT ELECTRODE. THE SPECIFIC AIMS ARE MOTIVATED BY OUR PRECLINICAL STUDIES THAT HAVE SHOWN THAT HYPOTHERMIA CAN PRESERVE RESIDUAL HEARING LONG-TERM POST-COCHLEAR IMPLANTATION. ENSURING THE SURVIVAL OF SENSITIVE HAIR CELLS AND NEURAL STRUCTURES IN THE COCHLEA WILL LEAD TO IMPROVED RESIDUAL HEARING PRESERVATION, WHICH IN THE LONG-TERM CAN IMPROVE SPEECH PERCEPTION OUTCOMES.

NON-INVASIVE SYSTEM TO DELIVER THERAPEUTIC HYPOTHERMIA FOR PROTECTION AGAINST NOISE-INDUCED HEARING LOSS - PROJECT SUMMARY RESTOREAR DEVICES LLC AIMS TO DEVELOP AND TEST APPLICATION OF MILD THERAPEUTIC HYPOTHERMIA TO MITIGATE NOISE- INDUCED HEARING LOSS (NIHL). NIHL IS GLOBAL PUBLIC HEALTH RISK AND IS AN IMPAIRMENT RESULTING FROM IRREVERSIBLE DAMAGE CAUSED TO THE SENSITIVE STRUCTURES AND NEURAL ELEMENTS IN THE COCHLEA. A CENTER FOR DISEASE CONTROL AND PREVENTION REPORT HIGHLIGHTS THAT 24% OF ADULTS AND 17% OF TEENAGERS IN THE UNITED STATES EXPERIENCE HEARING LOSS IN ONE OR BOTH EARS FROM EXPOSURE TO OCCUPATIONAL OR RECREATIONAL LOUD SOUND. NIHL HAS A HIGH PREVALENCE AMONG MEMBERS OF THE ARMED FORCES AND FIREFIGHTERS WHO ARE OCCUPATIONALLY AT-RISK. GLOBALLY, OCCUPATIONAL NOISE EXPOSURE IS RESPONSIBLE FOR 16% OF DISABLING HEARING LOSS (OVER 600 MILLION PEOPLE) WITH SIGNIFICANT LIFETIME COSTS PER PERSON IN THE US ALONE. PRELIMINARY AND PRIOR PUBLISHED RESULTS SHOW THAT CONTROLLED AND LOCALIZED THERAPEUTIC HYPOTHERMIA PROVIDED TO THE INNER EAR POST-NOISE OR SURGICAL TRAUMA CONSERVES SIGNIFICANT RESIDUAL HEARING AND PRESERVES SENSITIVE NEURAL STRUCTURES. OVER THE PAST 3 YEARS, THE UNIVERSITY OF MIAMI TEAM HAS ALSO SHOWN NOISE- INDUCED CHANGES IN HEARING AND BALANCE FUNCTIONS IN AT-RISK GROUPS OF FIREFIGHTERS. WITH THE PRIOR PHASE I SBIR AWARD, RESTOREAR DEVICES LLC DESIGNED, BUILT AND TESTED SYSTEMS AND PROTOCOLS TO DELIVER MILD THERAPEUTIC HYPOTHERMIA NON-INVASIVELY TO THE INNER EAR. WE MET ALL OF THE MILESTONES OUTLINED, INCLUDING TESTING THE EFFICACY OF OUR DEVICES FOR HUMAN APPLICATION USING COMPUTATIONAL MODELS OF HEAT TRANSFER AND STUDIES FROM CADAVERIC TEMPORAL BONES. WE ARE NOW READY TO TRANSLATE ONE SUCH DEVICE, REBOUNDTM, TO PRACTICE. THIS PHASE II SBIR PROPOSAL WILL TEST SAFETY AND EFFICACY OF REBOUND-DELIVERED MILD THERAPEUTIC HYPOTHERMIA TREATMENT IN MITIGATING NOISE-INDUCED HEARING LOSS IN FIREFIGHTERS. IN COLLABORATION WITH THE UNIVERSITY OF MIAMI AND SOUTH FLORIDA FIRE SERVICES, WE HAVE DESIGNED A CLINICAL STUDY TO TEST THIS THERAPY IN OCCUPATIONALLY AT-RISK FIREFIGHTERS. SUBJECTIVE AND FUNCTIONAL ASSESSMENTS REPEATED OVER TIME WILL ENABLE EVALUATION OF THE PROTECTIVE EFFECTS OF MILD THERAPEUTIC HYPOTHERMIA FOLLOWING NOISE EXPOSURE. RESTORE NYC WILL COLLABORATE WITH ITS TWO PARTNERS, THE MAYORS OFFICE TO END DOMESTIC AND GENDER-BASED VIOLENCE, AND QUEENS FAMILY JUSTICE CENTER, TO PROVIDE A HOLISTIC, VICTIM-CENTERED, AND MULTIDISCIPLINARY APPROACH TO MEET THE TRANSITIONAL HOUSING NEEDS IN THE COMMUNITY. IT IS ESTIMATED TO AFFECT BETWEEN 10 TO 20% OF ADULTS WORLDWIDE. WHILE MANY PATIENTS COPE WITH TINNITUS, NEARLY 1 IN 10 (OR MORE) EXPERIENCE SEVERE SYMPTOMS LEADING TO SIGNIFICANT DECREASE IN QUALITY OF LIFE AND SOCIOECONOMIC BURDEN TO THE SOCIETY. DESPITE SIGNIFICANT HEALTH BURDEN PRESENTED BY TINNITUS THERE EXISTS NO CURE AND THERE ARE NO SHORT TERM/ACUTE THERAPIES FOR REDUCING TINNITUS LOUDNESS. THE PROPOSED PROJECT AIMS TO TEST WHETHER THE NON- INVASIVE, NON-PHARMACEUTICAL DELIVERY OF MILD THERAPEUTIC HYPOTHERMIA (MTH) TARGETING MULTIPLE CELLULAR AND MOLECULAR PATHWAYS IN THE INNER EAR PROVIDES SYMPTOMATIC RELIEF OF TINNITUS AND REDUCES TINNITUS SEVERITY. RESTOREAR’S LONG-TERM GOALS ARE TO DEVELOP LOCALIZED THERAPEUTIC HYPOTHERMIA AS A NON-INVASIVE INTERVENTION FOR SYMPTOMATIC RELIEF OF TINNITUS AND TO MANAGE TINNITUS SEVERITY, PROVIDING ACUTE RELIEF TO PATIENTS WITH CHRONIC TINNITUS. HERE, THIS APPROACH WILL BE EXTENDED TO A GROUP OF PATIENTS SUFFERING FROM TINNITUS. WE WILL MEASURE THE EFFECTS OF THIS EXPERIMENTAL INTERVENTION USING STANDARD AND VALIDATED PSYCHOMETRIC AND AUDIOMETRIC FUNCTIONAL ASSESSMENTS. WITH STRONG RESULTS COLLECTED DURING THIS PHASE I PROJECT, WE AIM TO SHOW THAT MTH DELIVERED TO THE INNER EAR WILL MITIGATE TINNITUS SEVERITY AND LEAD TO A NEW APPROACH FOR ACUTE AND CHRONIC TINNITUS MANAGEMENT. RELEVANT TO THE RECENT CORONAVIRUS PANDEMIC, THE MAJORITY OF PATIENTS WITH SEVERE CORONAVIRUS DISEASE OF 2019 (COVID-19) DEVELOP ARDS AND NEED MECHANICAL VENTILATION AND ICU ADMISSION, PLACING SEVERE STRAINS ON THE MEDICAL SYSTEM. USE OF MECHANICAL VENTILATION (MV) DURING ARDS FOR CASES SUCH AS COVID-19 IS SUPPORTIVE UNTIL THE INFECTION HAS RESOLVED WHICH MAY TAKE UP TO 20 DAYS. WHILE MV IS USEFUL FOR VENTILATION AND SUPPORTING LIFE, IT CAUSES SEVERE ATROPHY OF RESPIRATORY MUSCLES THAT DELAYS EXTUBATION. FURTHERMORE, PROLONGED MV CAUSES SUBSTANTIAL MORBIDITY AND MORTALITY (I.E. LUNG INJURY, PNEUMONIA, LARYNGEAL INJURY, ETC.) THEREFORE, STRATEGIES TO CONDITION THE RESPIRATORY MUSCLES TO PREVENT ATROPHY DURING INTUBATION WILL LIKELY DECREASE MV TIME, THEREBY DECREASE MORBIDITY AND MORTALITY ASSOCIATED WITH MV, AND DECREASE VENTILATOR NEED AND CONSUMPTION OF VALUABLE RESOURCES. STRATEGIES HAVE BEEN DEVELOPED TO CONDITION OR ACTIVATE THE DIAPHRAGM MUSCLE DURING CHRONIC HIGH CERVICAL SPINAL CORD INJURY WITH IMPLANTS THAT STIMULATE THE PHRENIC NERVE OR DIAPHRAGM; HOWEVER, SUCH AN INVASIVE STRATEGY IS INCOMPATIBLE WITH THE ACUTE AND TEMPORARY NATURE OF ARDS. SCOTF WILL CREATE A PFS TASK FORCE WITH DATA-DRIVEN STRATEGIC AND ACTION PLAN THAT INCLUDES COMPREHENSIVE STRATEGIES TO ADDRESS SUBSTANCE USE DISORDER AND MENTAL HEALTH PROMOTI ON SERVICES OF THE PLAN, AS MEASURED BY ‘LIVING’ ACTION PLAN DOCUMENTATION. ADDITIONALLY EVIDENCE BASED PROGRAMS SUCH AS LIFE SKILLS, STRENGTHENING FAMILIES AND MENTAL HEALTH FIRST AID WILL BE IMPLEMENTED. FURTHERMORE, SCOTF PLANS TO INCORPORATE SCREENING, BRIEF INTERVENTION AND REFERRAL TO TREATMENT (SBIRT) INTO ROUTINE HEALTH SCREENINGS ADMINISTERED BY MEDICAL PROFESSIONALS AND DIVERSION COUNSELORS AS WELL AS PROVIDE SBIRT TRAINING TO MIDDLE AND HIGH SCHOOL STAFF. THE OVERALL GOAL IS TO IMPROVE YOUTH PERCEPTION OF HARM OF SUBSTANCE AND DECREASE PAST 30 DAY USE OF ALCOHOL AND SUBSTANCES.

ACCORDING TO THE IUCN CLASSIFICATIONS OF THREATENED AND ENDANGEREDSPECIES, AMPHIBIANS ARE THE MOST THREATENED GROUP OF VERTEBRATES, WITH 41 PERCENT OF SPECIESTHREATENED WITH EXTINCTION. FOR COMPARISON, THE SECOND MOST THREATENED GROUP IS MAMMALS WITH 27PERCENT OF SPECIES THREATENED. PEOPLE WORKING TO SAVE NATIVE AMPHIBIANS IN THE U.S. AND ITS TERRITORIESHAVE WITNESSED ENTIRE BREEDING POPULATIONS VANISH IN THE SPAN OF THEIR CAREERS. INNOVATIVE APPROACHESWILL TAKE TIME TO DEVELOP AND DEPLOY, DURING WHICH IMPORTANT ADAPTIVE GENETIC DIVERSITY WILL BE LOSTTHROUGH POPULATION DECLINE AND OR GENETIC DRIFT. BIOBANKING TECHNOLOGIES COULD SAFEGUARD AMPHIBIANGENETIC DIVERSITY FOR FUTURE CONSERVATION MANAGEMENT, BUT THE EXISTING TOOLKIT FOR AMPHIBIANS ISEXTREMELY LIMITED. OUR PROPOSED PROJECT AIMS TO DEVELOP NEW BIOBANKING METHODS TO ENHANCE RECOVERYAND MANAGEMENT OF THREATENED AMPHIBIANS, STARTING WITH THE WYOMING TOAD, ANAXYRUS BAXTERI, IS ONE OFNORTH AMERICAS MOST ENDANGERED AMPHIBIAN SPECIES. THE WYOMING TOADS RECOVERY PLAN IS THE ONLYAMPHIBIAN RECOVERY PLAN THAT LISTS CRYOPRESERVATION (BIOBANKING) COUPLED WITH REPRODUCTIVE TECHNOLOGIESAS A PRIORITY 1 ACTION: ACTION 4.3.1.5. PURSUE SPERM CRYOPRESERVATION AND IN VITRO FERTILIZATION (IVF). OURPROJECT AIMS TO FULFILL THIS ACTION BY EXPANDING CAPABILITIES BEYOND SPERM AND IVF. OUR GOALS ARE TO: (1)CRYOPRESERVE AND RECOVER LIVE EMBRYOS THAT DEVELOP INTO NEW REPRODUCTIVELY VIABLE INDIVIDUALS (2)GENERATE, CRYOPRESERVE, AND RECOVER EMBRYONIC CELL CULTURES (3) GENERATE, CRYOPRESERVE, AND RECOVERADULT SOMATIC CELL CULTURES FROM NON-LETHALLY COLLECTED TISSUE BIOPSIES AND (4) USE POPULATION GENOMICSTO IDENTIFY THE HIGHEST VALUE LINEAGES IN THE PEDIGREE FOR IMMEDIATE BIOBANKING AND CAPTIVE BREEDING.TOGETHER, THESE STRATEGIES WILL ENHANCE PRESENT AND FUTURE GENETIC MANAGEMENT EFFORTS FOR THE WYOMINGTOAD AND LAY THE GROUNDWORK FOR ADDITIONAL PRIORITY 1 RECOVERY ACTIONS, INCLUDING: 4.3.1.1. MAINTAINGENETIC DIVERSITY OF CAPTIVE POPULATION, 4.3.1.6. CONTINUE TO REDUCE DISEASE IN CAPTIVE TOADS, 4.3.2.1.CONTINUE GENETIC RESEARCH, 4.3.3.2. MINIMIZE SPREAD OF BD IN THE FIELD. THE ABILITY TO PRESERVE ANDRESTORE BOTH MALE AND FEMALE GENOMES FOR BREEDING, THROUGH THE GOALS WE HAVE PROPOSED HERE (LIVEEMBRYOS OR CELL LINES), WOULD FURTHER REDUCE RESOURCES AND COSTS TO ACHIEVE GENETIC MANAGEMENT GOALS.THE METHODS PROPOSED HERE FOR THE WYOMING TOAD MAY BE DIRECTLY TRANSFERABLE TO OTHER ENDANGEREDANAXYRUS SPECIES (SUCH AS THE HOUSTON TOAD, ARROYO TOAD, YOSEMITE TOAD, OR DIXIE VALLEY TOAD) AND, IFSUCCESSFUL, EMBRYO CRYOPRESERVATION SPECIFICALLY COULD BE BROADLY APPLIED TO ALL LISTED AMPHIBIAN SPECIES.WITH EFFECTIVE BIOBANKING TOOLS, THE WYOMING TOAD RECOVERY TEAM, AND OTHER AMPHIBIAN RECOVERY TEAMSCOULD IMMEDIATELY REVOLUTIONIZE RECOVERY STRATEGIES, SLOW FURTHER GENE DIVERSITY LOSS, ENABLE DISEASERESISTANCE RESEARCH, AND POTENTIALLY REDUCE OVERALL BREEDING PROGRAM COSTS.

SINCE OVER 40% OF AMERICANS LIVE IN COASTAL COUNTIES, IT IS ESSENTIAL THAT THE SUMMIT REFLECT THE SCOPE AND DIVERSITY OF THESE COMMUNITIES, AND THAT IT HELPS FOSTER RELATIONSHIPS THAT WILL MAKE RESTORATION MORE IMPACTFUL FOR ALL MEMBERS OF THESE COMMUNITIES. ACCORDING TO A STUDY BY GREEN 2.0, ETHNIC MINORITIES AND PEOPLE OF MULTI-RACIAL BACKGROUNDS COMPRISE ABOUT 38% OF THE U.S. POPULATION, BUT THE PERCENTAGE OF MINORITIES ON THE BOARDS OR GENERAL STAFF OF ENVIRONMENTAL ORGANIZATIONS DOES NOT EXCEED 16%. DESPITE THE ACCELERATING EXTINCTION CRISIS, FEWER THAN 50 TAXA (7%) OF U.S. THREATENED AND ENDANGERED ANIMALS HAD CELL LINES CRYOPRESERVED PRIOR TO THE LAUNCH OF OUR INFORMED BIOBANKING PROJECT PILOT IN DECEMBER 2022. TISSUE SAMPLES HAVE BEEN PRESERVED WITH VARIOUS METHODS, MANY OF WHICH HAVE YET TO BE VALIDATED, AND THEREFORE SOME TISSUES MAY NOT BE VIABLE FOR FUTURE APPLICATIONS. CELL LINES ARE THE GOLD STANDARD FOR BIOBANKING. THERE ARE UNIVERSAL METHODS FOR GENERATING CELL LINES FOR MAMMALS AND BIRDS FROM TISSUE BIOPSIES COLLECTED IN A NON-LETHAL MANNER. CRYOPRESERVATION AND THAW METHODS ARE UNIVERSAL, AND CELLLINES MAINTAIN THE HIGHEST VIABILITY RATES OF ANY BIOMATERIAL. OUR VISION WITH THE INFORMED BIOBANKING PROGRAM WAS TO DEVELOP A SCALABLE BIOBANKING WORKFLOW THAT ENSURES ALL U.S. ENDANGERED SPECIES CAN BE BIOBANKED AS PART OF LONG-TERM RECOVERY STRATEGIES. WE RECOGNIZE THIS IS AN AMBITIOUS ASPIRATION REQUIRING NEW SCIENCE AND TECHNOLOGY FOR THE FULL DIVERSITY OF ENDANGERED SPECIES, BUT FOR MAMMALS AND BIRDS BIOBANKING CELL LINES CAN BE CARRIED OUT NOW. FOR AMPHIBIANS AND REPTILES, TAXA FOR WHICH CELL LINES ARE DIFFICULT TO PRODUCE CURRENTLY, TISSUE SAMPLES CAN BE CRYOPRESERVED WITH THE BEST PRESENT DAY STANDARDIZED METHODS SO THAT IN THE FUTURE, WHEN THE SCIENCE HAS ADVANCED, CELL LINES CAN BE ESTABLISHED. PROJECT PURPOSE. OUR PURPOSE IS TO CONTINUE GENERATING CELL LINES AND BIOBANKING LIVING TISSUE SAMPLES FOR THREATENED AND ENDANGERED TAXA FOR USFWS REGION 2, INCLUSIVE OF AT-RISK AND PROPOSED LISTED TAXA OF MAJOR CONCERN TO THE REGION PROJECT OBJECTIVES. CELL LINES WILL BE STORED LONG-TERM AT THE USDA GERMPLASM BIOREPOSITORY. FOR AMPHIBIANS AND REPTILES, THE GOAL WILL BE TO BIOBANK LIVING TISSUES (AKA EXPRESS TISSUE BANKING) FOR 1 FEMALE AND 1 MALE, AND ALSO STORE THOSE TISSUES LONG-TERM AT THE USDA GERMPLASM BIOREPOSITORY. WE RECOGNIZE IT MAY NOT BE POSSIBLE TO COLLECT TISSUES FROM ALL THE TAXA LISTED IN TABLE 1 OVER THE COURSE OF A SINGLE YEAR DUE TO THE RARITY AND ELUSIVENESS OF SOME TAXA, AS WELL AS UNFORESEEN CIRCUMSTANCES THAT MAY IMPEDE COLLECTION (E.G. INCLEMENT WEATHER, WILDFIRE, ETC.). THEREFORE, OUR GOAL IS TO ESTABLISH CELL LINES FROM TISSUES ON A FIRST COME, FIRST SERVE BASIS UNTIL AVAILABLE FUNDS ARE EXHAUSTED. THE AIM OF ESTABLISHING CELL LINES AND EXPRESS TISSUE BANKING FOR ALL TAXA WILL BE A LONGER-TERM GOAL, DEPENDENT ON AVAILABLE FUNDS IN THE FOLLOWING YEARS. METHODOLOGY. SAMPLES WILL BE SHIPPED TO VIAGEN FOR CELL CULTURE GENERATION OR EXPRESS TISSUE BANKING. WHEN READY, SAMPLES WILL BE SHIPPED TO THE USDA GERMPLASM REPOSITORY FOR LONG-TERM STORAGE. REVIVE RESTORE WILL UPDATE USFWS STAFF THROUGHOUT THE PROCESS AS TO THE STATUS OF EACH TISSUE SAMPLE. TISSUE BIOPSIES WILL BE PURPOSELY COLLECTED FROM LIVE MAMMALS AND BIRDS DURING STANDARD FIELD MONITORING ACTIVITIES OR MEDICAL EXAMS PROCEDURES FOR CAPTIVE INDIVIDUALS. BIOBANKING EFFORTS CAN ALSO SALVAGE TISSUES OPPORTUNISTICALLY FROM INDIVIDUALS FOUND DECEASED IF THE CARCASS IS FOUND BEFORE THE TISSUES HAVE DEGRADED, OR FROM INDIVIDUALS TAKEN (EUTHANIZED) FOR PERMITTED RECOVERY ACTIVITIES OR OTHER JUSTIFIED CIRCUMSTANCES. SAMPLING OF REPTILES AND AMPHIBIANS WILL BE RESTRICTED TO POST-MORTEM TISSUE SAMPLING ADHERING TO THE CONDITIONS STATED ABOVE.

RESTORE AMERICA S ESTUARIES (RAE) WILL HOST THE 2025 LIVING SHORELINES TECH TRANSFER WORKSHOP (TTW) TO BRING TOGETHER THE COMMUNITY OF PRACTICE FOR KNOWLEDGE SHARING AND NETWORKING. ANTICIPATED TOPICS INCLUDE THE LATEST INNOVATIONS IN MATERIALS AND DESIGN, PROJECT CASE STUDIES AND LESSONS LEARNED, UPDATES ON THE POLICY AND REGULATORY ENVIRONMENT, AND OF COURSE FIELD TRIPS TO LOCAL SITES.AS WITH PREVIOUS WORKSHOPS, RAE WILL CO-HOST THIS EVENT WITH A LOCAL PARTNER AND WILL DESIGN THE CONTENT WITH THE HELP AND GUIDANCE OF A REGIONALLY-FOCUSED ADVISORY COMMITTEE. WE ANTICIPATE APPROXIMATELY 250 PARTICIPANTS TO ATTEND, REPRESENTING ALL ASPECTS OF THE FIELD, INCLUDING STATE AND FEDERAL GOVERNMENT, STATE AND LOCAL REGULATORY AGENCIES, ACADEMIA, PRIVATE SECTOR AND NON-PROFIT ORGANIZATIONS. THE EVENT TYPICALLY INCLUDES TWO FULL DAYS OF PROGRAMMING: A HALF-DAY FOR FIELD TRIPS TO LOCAL RESTORATION SITES, PLENARY SESSIONS, BREAKOUT DISCUSSIONS, AND A NETWORKING RECEPTION. TAKING PLACE OCTOBER 29-30, 2025 IN NEW HAVEN, CT, AND CO-HOSTED BY RAE AND OUR MEMBER GROUP SAVE THE SOUND, THIS WILL BE THE NATIONS LARGEST GATHERING FOCUSED ON THE SCIENCE, PRACTICE, AND POLICY OF LIVING SHORELINES AND NATURE-BASED SOLUTIONS. LIVING SHORELINES REFERS TO A VARIETY OF NATURE-BASED SHORELINE PROTECTION OPTIONS THAT ALLOW FOR NATURAL COASTAL PROCESSES TO REMAIN THROUGH THE STRATEGIC PLACEMENT OF PLANTS, STONE, SAND FILL, AND OTHER STRUCTURAL AND ORGANIC MATERIALS. LIVING SHORELINES BENEFIT COASTAL HABITATS, COMMUNITIES, AND ECOSYSTEMS ACROSS THE COUNTRY, AND THE COUNTLESS HUMANS AND SPECIES THAT LIVE WITHIN THEM. BENEFITS PROVIDED INCLUDE: 1. LIVING SHORELINES IMPROVE WATER QUALITY BY TRAPPING SEDIMENTS AND FILTERING POLLUTANTS. 2. LIVING SHORELINES PROVIDE SHALLOW WATER HABITAT AND A DIVERSITY OF PLANT SPECIES FOR AQUATIC AND TERRESTRIAL ANIMALS THEY ALSO PROVIDE SHADE TO KEEP WATER TEMPERATURES COOL, HELPING TO INCREASE OXYGEN LEVELS FOR FISH AND OTHER AQUATIC SPECIES. 3. LIVING SHORELINES REDUCE EROSION AND ABSORB WAVE ENERGY, GREATLY INCREASING SUSTAINABILITY AND RESISTANCE OF LOCAL COMMUNITIES AND ECOSYSTEMS TO WEATHER EVENTS AND OTHER NATURAL HAZARDS. 4.
